摘 要:目的建立适当的定性、定量检测方法,以期更加有效、准确地控制卷柏总黄酮的质量。方法采用HPLC法对卷柏总黄酮进行定性鉴别;以穗花杉双黄酮为对照,用UV法测定卷柏总黄酮的含量,HPLC法测定穗花杉双黄酮的含量;参照《中国药典》2015年版方法测定重金属及有害元素、有机氯农残和其他检查项。结果卷柏总黄酮和穗花杉双黄酮质量浓度分别在1.56~11.68、6.40~320.03μg·mL^(-1)范围内线性关系良好(r=1.0000),平均回收率分别为98.18%(RSD=1.76%)、96.66%(RSD=1.15%)。重金属及有害元素、有机氯农残和其他检查项均符合规定。结论所建立的方法简便、准确,指标成分科学有效,为该品种的质量评价与控制提供了有力的技术支持。Objective To establish reasonable qualitative and quantitative detection methods of Selaginella total flavones,which can be used to evaluate the quality effectively and accurately.Methods Selaginella total flavones were identified by HPLC.Amentoflavone as the standard,HPLC method was employed to determine the content of amentoflavone and UV method was used to determine the content of Selaginella total flavones.According to the methods recorded in Chinese Pharmacopoeia(2015 edition),heavy metals and harmful elements,organochlorine pesticide residues,and other requirementitems were studied.Results The Selaginella total flavones showed a good linear relationship in the range of 1.56-11.68μg·mL^(-1)(r1=1.0000).The average recovery was 98.18%with RSD 1.76%.Amentoflavone showed a good linear relationship in the range of 6.40-320.03μg·mL^(-1)(r2=1.0000).The average recovery was 96.66%with RSD 1.15%.Heavy metals and harmful elements,organochlorine pesticide residues,and other requirements were all complied with the specification.Conclusion The methods established in the study were simple and accurate,and the chemical composition makers were effective.This paper provides a strong technical support for the quality evaluation and control of Selaginella total flavones.
