摘 要:目的采用代谢组学的方法研究青蒿鳖甲汤对肝细胞癌模型小鼠血浆代谢物的影响,由筛选出的差异代谢物来探究其相关作用机制。方法注射鼠源肝癌细胞株H22细胞对小鼠进行造模,随机分为正常对照组、模型组和青蒿鳖甲汤组,运用UHPLC-Q-Exactive液质联用技术检测各组小鼠血浆中的代谢物变化并进行多元统计分析研究,筛选出差异性代谢物及对应的相关代谢通路。结果多元分析结果显示,3组的代谢物存在显著差异,寻找出肝癌模型组及给予青蒿鳖甲汤干预后模型小鼠的差异代谢物15个,涉及31条相关的代谢通路,经通路富集分析发现,重要性得分在前三的代谢通路分别是胆汁分泌、鞘脂代谢、类固醇激素生物合成。结论青蒿鳖甲汤能有效改善部分紊乱的代谢物水平,为进一步对青蒿鳖甲汤改善肝癌及后期发热的作用机制研究提供理论依据。Objective To investigate the effect of Qinghao Biejia Decoction on plasma metabolites in hepatocellular carcinoma model mice by metabolomics, and to investigate the related mechanism of action by the screened differential metabolites. Methods The mice were randomly divided into normal control group, model group and Qinghao Biejia Decoction group by injection of mouse hepatoma cell line H22 cells to model mice. UHPLC-Q Exactive was used to detect the metabolic changes in the plasma of mice in each group, and multivariate statistical analysis was conducted to screen out the differential metabolites and the corresponding related metabolic pathways. Results Multivariate analysis showed that three groups of metabolites were stored in the significant difference, 15 metabolites were found in liver cancer model and treated with Qinghao Biejia Decoction, involving 31 related metabolic pathways. Conclusion Qinghao Biejia Decoction can effectively improve the level of metabolites in some disorders, and provide a theoretical basis for further study on the mechanism of improving liver cancer and later fever.
