摘 要:由大丽轮枝菌(Verticillium dahliae)引起的棉花黄萎病每年给我国农业生产造成严重经济损失。为了明确不同抗性陆地棉(Gossypium hirsutum)品种中病程相关基因(GhPR基因)响应不同致病型大丽轮枝菌的表达差异,揭示GhPR基因在棉花与大丽轮枝菌互作过程中的免疫反应变化,本研究用寄主来源的棉花黄萎病菌落叶型菌株V592和非落叶型菌株I6以及非寄主来源的落叶型菌株S1和非落叶型菌株V31分别对2个感病棉花品种和1个抗病棉花品种的无菌根系进行诱导处理,检测GhPR基因的表达情况。结果显示,棉花黄萎病菌落叶型菌株V592和非落叶型菌株I6均能诱导3个棉花品种的GhPR基因普遍上调表达;在感病品种军棉1号上,2个菌株诱导Gh-PR基因达到最大相对表达量的基因个数相当;在感病品种新陆早1号上,I6菌株诱导GhPR基因达到最大相对表达量的基因个数比V592菌株多;表明在棉花感病品种上,非落叶型菌株诱导GhPR基因上调表达的能力与落叶型菌株的诱导能力相当,甚至更强。在抗病品种中植棉2号上,落叶型菌株V592诱导GhPR基因达到最大相对表达量的基因个数及上调表达量均显著高于I6菌株诱导的上调表达量,表明在棉花抗病品种上,落叶型菌株诱导GhPR基因上调表达的能力比非落叶型菌株的诱导能力强。对非寄主来源的2个大丽轮枝菌菌株,非落叶型菌株V31诱导感病棉花品种GhPR基因达到最大相对表达量的基因个数比落叶型菌株S1多,落叶型菌株S1诱导抗病棉花品种GhPR基因达到最大相对表达量的基因个数比非落叶型菌株V31多。结果表明,棉花感病品种的GhPR基因对落叶型和非落叶型大丽轮枝菌侵染均有一定的响应能力,棉花抗病品种的GhPR基因对落叶型菌系侵染具有更强的响应能力。Verticillium wilt disease becomes a major threat to many economically important crops.In order to determine the expression differences of pathogenesis-related genes(GhPR gene)in Gossypium hirsutum cultivars in response to different pathotype of Verticillium dahliae,and to reveal the changes of GhPR gene in the immune response of cotton,we treated the roots of two susceptible cultivars and one resistant cultivar of G.hirsutum with defoliating strain V592 and non-defoliating strain I6 isolated from upland cotton,and defoliating strain S1 and non-defoliating strain V31 isolated from other plants,respectively;then transcriptional expression of GhPR were measured.The results showed that the defoliating strains V592 and non-defoliating strain I6 from cotton could induce the up-regulated expressions of different GhPR genes in three G.hirsutum cultivars.In susceptible cultivar Junmian 1,the number of GhPR genes with the maximum relative expression induced by V592 strain was similar to that induced by I6 strain;in susceptible cultivar Xinluzao 1,the number of Gh-PR genes with the maximum relative expression induced by I6 strain was higher than that induced by V592 strain.It suggested that in susceptible cultivars,non-defoliating strain is similar or more effective to trigger GhPR gene expressions compare to defoliating strain.In resistant cultivar Zhongzhimian 2,the number of GhPR genes with the maximum relative expression and the transcriptional level of Gh-PR genes induced by V592 strain were significantly higher than those induced by I6 strain.It indicated that defoliating strain is more effective to trigger GhPR gene expressions than non-defoliating strain in resistant cultivar.For two strains of V.dahliae isolated from other plants,in susceptible cultivars,the number of GhPR genes with the maximum relative expression induced by non-defoliating V31 was higher than that induced by defoliating strain S1;in resistant cultivar Zhongzhimian 2,the number of GhPR genes with the maximum relative expression induced by S1 strai
