摘  要:目的构建绞股蓝皂苷XLⅨ纳米结构脂质载体(XLⅨ-NLCs),探讨其对糖尿病足(DF)大鼠的创面愈合以及对内皮素(ET)和降钙素基因相关肽(CGRP)平衡的影响。方法利用高压均质法制备XLⅨ-NLCs,培养表皮干细胞(ESCs),并利用链脲佐菌素和足背部创伤构建大鼠糖尿病足创伤模型。ESCs细胞分为4组:对照组,无处理的ESCs;NLCs组,经NLCs处理的ESCs;XLⅨ组,经XLⅨ处理的ESCs;XLⅨ-NLCs组,经XLⅨ-NLCs处理的ESCs。使用MTT法检测细胞活性。大鼠分为正常组(n=20)、DF组(n=20)、DF+NLCs组(n=20)、DF+XLⅨ组(n=20)、DF+XLⅨ-NLCs组(n=20)共5组。测定各组大鼠创面大小,使用HE染色检测大鼠创面组织的变化。使用试剂盒法检测大鼠血液中ET和CGRP的水平。结果与其他各组相比,XLⅨ-NLCs组的ESCs细胞活力最好(P<0.05)。伤后7 d时,与DF组、DF+NLCs组、DF+XLⅨ组相比,DF+XLⅨ-NLCs组创面修复最好,血液ET水平最低,且CGRP水平最高(P<0.05)。结论XLⅨ-NLCs可增强表皮干细胞活性,加快DF大鼠的创面修复,降低DF大鼠的ET水平并增加CGRP的水平。Objective To construct gynostemma saponin XLⅨnanostructured lipid carriers(XLⅨ-NLCs),and to explore its regulation on wound healing in diabetic foot(DF)rats and its effect on the balance of endothelin ET and calcitonin gene-related peptide CGRP.Methods XLⅨ-NLCs were prepared by high-pressure homogenization,epidermal stem cells(ESCs)were cultured,and DF rats were constructed using streptozotocin and dorsal trauma.ESCs cells were divided into 4 groups:control group,untreated ESCs;NLCs group,NLCs treated ESCs;XLⅨgroup,XLⅨtreated ESCs;XLⅨ-NLCs group,XLⅨ-NLCs treated ESCs.MTT method was used to detect cell activity.Rats were divided into normal group,DF group,DF+NLCs group,DF+XLⅨgroup,DF+XLⅨ-NLCs group for a total of 5 groups.The size of the rats'wounds in each group was measured,and HE staining was used to detect changes in the tissue of the rats'wounds.The levels of ET and CGRP in the blood of rats were detected using the kit method.Results Compared with other groups,XLⅨ-NLCs group showed the best cell activity of ESCs(P<0.05).Compared with DF group,DF+NLCs group and DF+XLⅨgroup,DF+XLⅨ-NLCs group had the best wound healing,the lowest blood ET level,and the highest CGRP level 7 days after operation(P<0.05).Conclusion XLⅨ-NLCs can enhance epidermal stem cell activity,accelerate wound healing in DF rats,reduce ET levels in DF rats,and increase CGRP levels.
