摘 要:目的探讨人参皂甙Rbl(ginsenosides Rbl,Gs-Rb1)改善心力衰竭(heart failure,HF)是否与心肌细胞的钙调控有关。方法将阿霉素诱导的HF大鼠随机分为HF组(n=16)和Gs-Rbl组[70 mg/(kg·d),n=18],另随机选取同龄大鼠作为对照组(n=10)。干预结束并完成心脏超声后取大鼠血和摘除心脏分离心肌细胞,测定氨基末端脑钠肽前体(N-terminal pro-brain natriuretic peptide,NT-proBNP)浓度以及检测细胞内钙瞬变、肌质网内钙容量和线粒体的Ca2+摄取和释放。结果Gs-Rb1显著提高HF大鼠左心室射血分数(left ventricular ejection fraction,LVEF)和降低血清NT-proBNP浓度;Gs-Rb1显著改善HF大鼠心肌细胞动作电位过程中的钙火花频率和钙瞬变峰值、心肌细胞Fluo-5N/AM染色后的荧光强度和荧光强度变化平均值以及心肌细胞线粒体的Ca2+摄取和释放等。结论Gs-Rb1可能通过调控心肌细胞水平、肌质网和线粒体等的Ca2+“交流”改善心力衰竭。Objectives The present study elucidated whether the effect of Ginsenosides-Rbl(Gs-Rb1)improving rat cardiac function with heart failure(HF)was performed by calcium regulation in cell level and sub-cell level in cardiomyocytes.Methods Adriamycin-induced rats with HF were randomly divided into HF group(n=16)and Gs-Rb1 group[70 mg/(kg·d),n=18],and the health age-matched rat was as control group(n=10).After the intervention being performed,the left ventricular ejection fraction(LVEF)was analyzed by echoeardiography.Afterward,blood and heart for ventricular cardiomyocytes were rapidly acquired.Concentration of N-terminal pro-brain natriuretic peptide(NT-proBNP)was measured,and Ca2+transients in cardiomyocyte,Ca2+contents in sarcoplasmic reticulum,Ca2+uptake and Ca2+efflux in mitochondrial were assayed by laser scanning confocal microscope.Results Gs-Rb1 significantly increased LVEF and significantly decreased concentration of NT-proBNP.Gs-Rb1 significantly improved both Ca2+spark frequency and calcium transient peak during action potential process,the fluorescence intensity and its fluorescence averages in cardiomyocytes with SR-entrapped fluo-5N/AM,and both Ca2+uptaked and Ca2+efflux in mitochondria of cardiomyocytes in rat with HF.Conclusions Gs-Rb1 improving HF was partly fulfilled through adjusting Ca2+level in cardiomyocyte,sarcoplasmic reticulum and mitochondrial at least.
