摘  要:利用1 000 L工业级别发酵罐规模化制备赤芝液态深层发酵活性产物,获得的30 L发酵产物经醇提浓缩后用石油醚、乙酸乙酯依次萃取,得到3种不同极性部位的萃取物,分别评价其抗肿瘤活性。结果表明:发酵过程中添加油酸可以促进活性产物的合成,发酵产物中三萜及甾醇含量高达8.91 mg∕(100 mg干菌丝);石油醚萃取物与乙酸乙酯萃取物均表现出良好的抗L1210和K562肿瘤细胞活性。可见,液态深层发酵技术是规模化制备赤芝活性物质的有效手段,可为后续活性成分分离纯化的研究提供稳定的材料来源。The 30 L fermentation product of Ganoderma lucidum was prepared by large-scale production of 1000 L industrial grade fermentor.After ethanol extraction and concentration,the fermentation product was extracted with petroleum ether and ethyl acetate in turn to obtain three extracts with different polarity,and their antitumor activities were evaluated respectively.The results showed that the addition of oleic acid could promote the synthesis of active product,and the content of triterpenoids and sterols in fermentation product was up to 8.91 mg∕(100 mg dry mycelium);Petroleum ether extract and ethyl acetate extract showed good anti-L1210 and K562 tumor activity.It can be seen that submerged fermentation technology is an effective method for large-scale preparation of active substances of G.lucidum,which can provide a stable material source for the subsequent study of separation and purification of active ingredients.
