摘  要:2020年5月,在上海市浦东新区和青浦区设施栽培黄瓜上发现一种能导致黄瓜茎基部腐烂并伴有茎秆流胶、最终整株死亡症状的病害。经病菌分离、致病性测定及分子生物学鉴定表明:病原菌分离物在NA培养基上菌落呈圆形,灰白色,有很浓烈的臭味,无荧光反应,革兰氏染色呈阴性;经BLASTn比对,其16S rRNA序列和RNA聚合酶β基因(rpoB)序列与胡萝卜软腐果胶杆菌巴西亚种的相似性分别为99.86%和97.14%;基于16S rRNA序列构建系统发育进化树,发现其在系统发育树上与胡萝卜软腐果胶杆菌巴西亚种的类群聚在一族;又经特异性引物鉴定,确定该分离物为胡萝卜软腐果胶杆菌巴西亚种(Pectobacterium carotovorum subsp.brasiliense)。In May 2020,a bacterial disease was found on cucumber cultivated in facilities in Pudong New District and Qingpu District of Shanghai,which could cause the stem rot,stem gumming and eventually the whole plant death.The results of pathogen isolation,pathogenicity test and molecular biological identification showed that the colony of pathogen isolate on NA medium was round,gray white,with strong odor,no fluorescence reaction and gram negative staining.The similarity of 16S rRNA sequence and RNA polymeraseβgene sequence between the pathogen and Pectobacterium carotovorum subsp.brasiliense(Pcb)was 99.86%and 97.14%respectively by BLASTn.Based on the 16S rRNA sequence,the phylogenetic tree was constructed,and it was found that the pathogen was clustered with Pcb;The isolate was also identified as Pcb by specific primers.
