摘 要:目的明确异丙酚对脓毒症小鼠模型中性粒细胞胞外诱捕网(NETs)生成和炎症因子表达的影响。方法雄性8~10周龄C57BL/6小鼠40只,按照随机数字表法分为对照组、脂多糖(LPS)组、异丙酚组和脱氧核糖核酸酶(DNase)组,每组各10只。腹腔注射LPS建立脓毒症模型;异丙酚组小鼠腹腔注射异丙酚全身麻醉1 h;DNase组腹腔注射DNase维持1 h;对照组腹腔注射等量的0.9%氯化钠溶液,观察相同时间。PicoGreen荧光染料法检测4组小鼠血浆游离DNA(cf-DNA/NETs)水平;酶联免疫吸附试验测定Toll样受体-4(TLR-4)、肿瘤坏死因子-α(TNF-α)和髓过氧化物酶(MPO)水平。结果LPS组小鼠的cf-DNA/NETs、TLR-4、TNF-α和MPO水平分别为(921.23±27.14)ng/mL、(13.88±1.12)ng/mL、(10253.14±199.25)pg/mL、(135.67±9.87)milliunits/mL,均明显高于对照组[(166.54±11.83)、(2.21±0.29)ng/mL、(2924.68±25.53)pg/mL、(32.83±1.47)milliunits/mL],差异均有统计学意义(P<0.05);异丙酚组小鼠的cf-DNA/NETs、TLR-4、TNF-α和MPO水平[(611.55±35.17)ng/mL、(9.97±0.51)ng/mL、(6505.03±79.76)pg/mL、(87.77±6.06)milliunits/mL]较LPS组明显降低,差异均有统计学意义(P<0.05);DNase组小鼠的TLR-4、TNF-α水平[(5.39±0.28)ng/mL、(4702.74±114.21)pg/mL]高于对照组,差异均有统计学意义(P<0.05);DNase组小鼠的cf-DNA/NETs、MPO水平[(203.52±7.01)ng/mL,(41.89±3.84)milliunits/mL]与对照组相比,差异均无统计学意义(P>0.05)。结论异丙酚对脓毒症小鼠模型NETs的生成和炎症因子表达具有一定的抑制作用,可适用于脓毒症患者的临床麻醉。Objective To investigate the effect of Propofol on production of neutrophil extracellular traps(NETs)and expression of inflammatory cytokines in sepsis mice.Methods Forty male C57BL/6 mice aged 8-10 weeks were randomly divided into control group,lipopolysaccharide(LPS)group,Propofol group and deoxyribonuclease(DNase)group,10 cases in each group.Sepsis model was established by intraperitoneal injection of LPS;the Propofol proup were given general anesthesia with intraperitoneal injection propofol for 1 hour;the DNase group were intraperitoneal injected DNase,then maintain for 1 hour,the control group was injected with the same amount of physiological saline maintained the same time.PicoGreen fluorescent dyes was used to detect plasma circulating free DNA cf-DNA/NETs concentration;and enzyme linked immunosorbent assay(ELISA)was used to assess the concentration of Toll like receptor-4(TLR-4),tumor necrosis factor-α(TNF-α)and myeloperoxidase(MPO)in 4 groups.Results The levels of cf-DNA/NETs,TLR-4,TNF-αand MPO in the LPS group were(921.23±27.14)ng/mL,(13.88±1.12)ng/mL,(10253.14±199.25)pg/mL,(135.67±9.87)milliunits/mL,which were significantly higher than those in the control group[(166.54±11.83),(2.21±0.29)ng/mL,(2924.68±25.53)pg/mL,(32.83±1.47)milliunits/mL],the differences were statistically significant(P<0.05);the levels of cf-DNA/NETs,TLR-4,TNF-αand MPO in the propofol group[(611.55±35.17)ng/mL,(9.97±0.51)ng/mL,(6505.03±79.76)pg/mL,(87.77±6.06)milliunits/mL]were significantly lower than those in the LPS group,and the difference was statistically significant(P<0.05);the levels of TLR-4 and TNF-αin the DNase group[(5.39±0.28)ng/mL,(4702.74±114.21)pg/mL]were higher than those in the control group,the differences were statistically significant(P<0.05);Compared with the control group,the cf-DNA/NETs and MPO levels of mice in the DNase group[(203.52±7.01)ng/mL,(41.89±3.84)milliunits/mL],the differences were not statistically significant(P>0.05).Conclusion Propofol can inhibit the production of NETs a
