摘  要:目的研究川芎嗪减轻大鼠视网膜缺血-再灌注损伤(RIRI)的作用及可能的分子机制。方法成年雄性SD大鼠48只按照随机数字表法分为对照组、RIRI组、川芎嗪组,每组各16只。RIRI组、川芎嗪组采用高眼压法建立RIRI模型,川芎嗪组造模前30 min给予盐酸川芎嗪注射液干预,RIRI组和对照组给予0.9%氯化钠溶液腹腔注射。造模后72 h,苏木精-伊红染色观察视网膜损伤情况,酶联免疫吸附试验法检测视网膜中白细胞介素(IL)-17、IL-23的含量,蛋白质印迹法检测视网膜中Bax、Bcl-2、Caspase-3的表达量。结果与对照组比较,RIRI组视网膜厚度变薄[(104.52±17.78)μm],视网膜神经节细胞数目及Bcl-2的表达量[(376.68±76.67)个/mm^(2)、0.39±0.08]减少,视网膜中IL-17、IL-23的含量[(3.28±0.96)、(2.89±0.64) pg/mL]及Bax、Caspase-3的表达量(0.92±0.24、0.73±0.20)增加,差异均有统计学意义(P <0.05);与RIRI组比较,川芎嗪组视网膜厚度增厚[(122.27±22.27)μm],视网膜神经节细胞数目及Bcl-2的表达量[(484.37±71.47)个/mm^(2)、0.70±0.16]增加,视网膜中IL-17、IL-23的含量[(1.94±0.35)、(1.34±0.28) pg/mL]及Bax、Caspase-3的表达量(0.35±0.10、0.50±0.09)减少,差异均有统计学意义(P <0.05)。结论川芎嗪能够减轻大鼠RIRI且这一作用与抗炎及抗凋亡有关。Objective To study the effect of ligustrazine on reduing retinal ischemia-reperfusion injury(RIRI) in rats based on anti-inflammatory and anti apoptosis effects. Methods A total of 48 adult male SD rats were randomly divided into control group,RIRI group and ligustrazine group,16 rats in each group. RIRI group and ligustrazine group were used to establish RIRI model by high intraocular pressure method. Ligustrazine group were intervened by Ligustrazine Hydrochloride Injection 30 minutes before modeling,RIRI group and control group were given intraperitoneal injection of 0. 9% sodium chloride solution. Seventy-two hours after the establishment of the model,Hematoxylin-eosin staining was used to observe the damage of retina,the contents of interleukin(IL)-17 and IL-23 in retina were detected by enzyme-linked immunosorbent assay,and the expressions of Bax,Bcl-2 and Caspase-3 in retina were detected by Western blotting. Results Compared with the control group,the retinal thickness of the RIRI group became thinner [(104. 52 ± 17. 78) μm],the number of retinal ganglion cells and the expression of Bcl-2 [(376. 68 ± 76. 67)/mm^(2),0. 39 ± 0. 08]decreased,and the content of IL-17 and IL-23 in the retina [(3. 28 ± 0. 96),(2. 89 ±0. 64) pg/m L] and the expression of Bax and Caspase-3(0. 92 ± 0. 24,0. 73 ± 0. 20) increased,and the differences were statistically significant(P < 0. 05);Compared with the RIRI group,the retinal thickness in the ligustrazine group increased [(122. 27 ± 22. 27) μm],the number of retinal ganglion cells and the expression of Bcl-2[(484. 37 ± 71. 47)/mm^(2),0. 70 ± 0. 16],the content of IL-17 and IL-23 in the retina [(1. 94 ± 0. 35),(1. 34 ± 0. 28) pg/m L] and the expression of Bax and Caspase-3(0. 35 ± 0. 10,0. 50 ± 0. 09) decreased,and the differences were statistically significant(P < 0. 05). Conclusion Ligustrazine can reduce RIRI in rats,which is related to anti-inflammation and anti-apoptosis.
