摘  要:目的研究布托啡诺通过蛋白激酶B(AKT)信号通路对大鼠心肌缺血再灌注损伤的影响。方法选用Wistar雄性大鼠,结扎左冠状动脉前降支(LAD)建立大鼠心肌缺血再灌注模型,并随机分为假手术组、模型组和实验组,每组各10只,另取10只不结扎LAD的大鼠设为假手术组。实验组大鼠静脉注射布托啡诺50μg·kg^(-1),模型组和假手术组大鼠静脉注射10 m L·kg^(-1)生理盐水;以酶联免疫吸附法(ELISA)试剂盒测定肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)含量以及心肌肌钙蛋白Ⅰ(CTnⅠ)、肌酸激酶同工酶(CK-MB);以蛋白质印迹(Western blot)法检测AKT的表达。结果假手术组、模型组和实验组大鼠TNF-α分别为(45.21±4.87),(135.83±7.28),(95.31±5.91)ng·L^(-1);IL-6分别为(55.31±5.14),(174.35±6.33),(137.32±5.24)ng·L^(-1);CTnⅠ分别为(0.84±0.16),(3.11±0.56),(1.54±0.21)ng·m L^(-1);CK-MB分别为(206.41±45.14),(1044.51±64.25),(519.33±63.21)U·L^(-1);AKT的表达分别为1.00±0.11,0.42±0.04,0.78±0.01,上述指标中假手术组与模型组比较,差异均有统计学意义(均P<0.05);模型组和实验组比较,差异均有统计学意义(均P<0.05)。结论布托啡诺可通过调节AKT表达,有效降低炎性因子水平和增强心肌功能,从而改善心肌缺血再灌注损伤。ObjectiveTo explore the effect of butorphanol on myocardial ischemia-reperfusion injury in rats through the protein kinase B(AKT)signaling pathway.Methods Male Wistar rats were selected,and the left anterior descending(LAD)coronary artery was ligated to establish a rat model of myocardial ischemia-reperfusion.The rats were divided randomly into sham operation group,model group and test group,with 10 rats in each group.Another 10 rats without LAD ligation were set as sham operation group.Rats in test group were intravenously injected with 50μg·kg^(-1)butorphanol.Rats in model group and sham operation group were intravenously injected with 10 m L·kg^(-1)normal saline.Enzyme-linked immunosorbent assay(ELISA)kit was used to detect the contents of tumor necrosis factor-α(TNF-α)and interleukin-6(IL-6),and the levels of cardiac troponinⅠ(CTnⅠ)and creatine kinase isoenzyme(CK-MB).Western blot was performed to detect the expression of AKT.Results TNF-αcontents of rats in sham operation group,model group and test group were(45.21±4.87),(135.83±7.28)and(95.31±5.91)ng·L^(-1),IL-6 contents were(55.31±5.14),(174.35±6.33)and(137.32±5.24)ng·L^(-1),CTnⅠlevels were(0.84±0.16),(3.11±0.56)and(1.54±0.21)ng·m L^(-1),respectively;CK-MB levels were(206.41±45.14),(1044.51±64.25)and(519.33±63.21)U·L^(-1),the expressions of AKT were1.00±0.11,0.42±0.04,and 0.78±0.01.In terms of the above indicators,the differences between sham operation group and model group were statistically significant(all P<0.05);the differences between model group and test group were statistically significant(all P<0.05).Conclusion Butorphanol can effectively reduce the levels of inflammatory factors and enhance myocardial function by regulating the expression of AKT,thereby alleviating myocardial ischemiareperfusion injury.
