健康人群中IgG类血型抗体高低效价间的肠道微生物差异

摘　　要：目的:探讨影响健康人群IgG类抗体效价水平的关键菌群。方法:2020年1月至2020年2月招募43例复旦大学在校学生为健康志愿者,收集静脉血和新鲜粪便样本。采用抗人球蛋白凝胶卡测定IgG类血型抗体效价,以抗体效价1∶128为临界点分为IgG类抗A抗体效价低水平组(OALG,≤128)、IgG类抗A抗体效价高水平组(OAHG,≥256)、IgG类抗B抗体效价低水平组(OBLG,≤128)和IgG类抗B抗体效价高水平组(OBHG,≥256)。将O型血样本进行16S rRNA V3-4区域测序分析,对肠道菌群进行α多样性、β多样性及线性判别分析。结果:43例志愿者中A型血12例,B型血17例,未发现抗体效价≥256的样本。O型血14例,抗体效价≥256的样本8例。血型效价高低组间(OAHG vs OALG、OBHG vs OBLG)在属水平上Shannon指数差异无统计学意义(P=0.4111、0.2278),而菌群内某些菌群相对丰度差异有统计学意义(P<0.05)。LDA值大于2.0时,OAHG和OALG有17种菌群有差异,其中OAHG中LDA最大值的菌群是普通拟杆菌,OBHG和OBLG有13种菌群有差异,其中OBHG中LDA最大值的菌群是毛螺菌科。结论:O型血中,血型抗体高低效价间,在属水平上肠道微生物菌群多样性差异无统计学意义,但在某些菌群的相对丰度上存在显著性差异。Objective:To explore the key flora affecting the blood group IgG antibody titer.Methods:Totally,43 students from Fudan University were recruited as healthy volunteers and their venous blood and fresh stool samples were collected from January to February 2020.Anti-human globulin gel card was used to determine the IgG antibody titers,and participants were divided into high-level and low-level groups with a cutoff point of 1∶128,low IgG class anti-A antibody titer group(OALG,≤128),high IgG anti-A antibody titer group(OAHG,≥256),low IgG class anti-B antibody titer group(OBLG,≤128),high IgG class anti-B antibody titer group(OBHG,≥256).O-type blood samples were analyzed by 16S rRNA V3-4 regional sequencing analysis,andαdiversity,βdiversity,and Linear discriminant analysis Effect Size(LEfSe)analyses were perfomed.Results:Among the paticipants,12 were blood type A and 17 were blood type B.No samples with antibody titer≥256 were found.There were 14 cases of blood type O,and 8 cases of samples with antibody titer≥256.At the genus level,there was no statistically significant difference in Shannon index(P=0.4111,0.2278),but the relative abundance of a certain flora was significantly different between the two groups(OAHG vs OALG,OBHG vs OBLG).When the LDA value is greater than 2.0,there are 17 kinds of flora(OAHG vs OALG),and 13 kinds of flora(OBHG vs OBLG).In the OAHG group,the bacterial group with the largest LDA value is Bacteroides vulgaris and in the OBHG group,the bacterial group with the largest value of LDA is Lacetospiraceae.Conclusions:In blood type O,when comparing the high and low IgG antibody titer groups,there is no statistical significant difference in the bacterial diversity of intestinal flora at the genus level,but there are significant differences in the relative abundance of certain flora.
