摘  要:探讨山楂酸对脂多糖诱导的RAW264.7细胞炎症反应的保护作用及其对信号转导和转录活化因子3(STAT3)磷酸化的抑制作用。采用Griess法检测一氧化氮(NO)的生成,ELISA法检测细胞中白介素6(IL-6)、白介素1β(IL-1β)和白介素10(IL-10)的释放量;Western blot法检测诱导型一氧化氮合酶(iNOS)、磷酸化信号转导和转录活化因子3(p-STAT3)、磷酸化Janus激酶2(p-JAK2)和细胞因子信号抑制物3(SOCS3)蛋白的表达,DCFH-DA荧光探针法测定细胞中的活性氧(ROS)水平。结果表明:山楂酸可显著抑制脂多糖诱导的RAW264.7细胞NO的生成和ROS的产生,降低白介素6和白介素1β的释放量,提高白介素10的含量;山楂酸对脂多糖诱导的RAW264.7细胞诱导型一氧化氮合酶的表达具有明显的抑制作用,进一步的研究表明,山楂酸呈剂量和时间依赖性地抑制LPS诱导的RAW264.7细胞信号转导和转录活化因子3的磷酸化并抑制其核转位,并能抑制Janus激酶2的活化和激活细胞因子信号抑制物3蛋白的表达。山楂酸对脂多糖诱导的RAW264.7细胞炎症反应有保护作用,其机制与抑制信号转导和转录活化因子3的磷酸化相关。In the paper,the protective effect of maslinic acid(MA)on LPS-induced inflammatory response in RAW264.7 cells was investigated,and its inhibition of transcription 3(STAT3)phosphorylation,a signal transducer and activator,was also explored.NO generation was measured by Griess method.The levels of IL-6,IL-1βand IL-10 were detected by ELISA.The relative expression of iNOS,p-STAT3,p-JAK2 and SOCS3 were determined by Western blot.The release of reactive oxygen(ROS)was evaluated using DCFH-DA fluorescence.Results showed that MA could significantly inhibit the NO generation and the production of ROS.MA also decreased the releases of IL-6 and IL-1β,increased the content of IL-10.The expression of iNOS in LPS-induced RAW264.7 cells was inhibited by MA.In addition,the phosphorylation and the nuclear translocation of STAT3 could be suppressed by MA in dose and time-dependent manner.The phosphorylation of JAK2 was also inhibited by MA,while the expression of SOCS3 could be activated.These results suggested that MA protected against LPS-induced inflammatory response in RAW264.7 cells,and its mechanism was probably associated with the inhibition of the phosphorylation of STAT3.
