摘 要:目的:研究金莲花中5个黄酮类化合物的体内代谢产物。方法:将荭草素、荭草素-2″-O-β-L-半乳糖苷、牡荆素、2″-O-(2-甲基丁酰基)牡荆素、异槲皮素分别以静脉注射的方式给予大鼠,给药10 min后取血,利用高效液相色谱串联质谱法(HPLC-MS/MS)对血浆中的代谢产物进行分析。结果:大鼠体内共检测到20个代谢产物,其中5个来源于荭草素、2个来源于荭草素-2″-O-β-L-半乳糖苷、3个来源于牡荆素、5个来源于2″-O-(2-甲基丁酰基)牡荆素、5个来源于异槲皮素。通过准分子离子峰及二级碎片推测20个代谢产物的结构及5个原型化合物的代谢规律。结论:5个黄酮类成分均可以通过氧化、甲基化、硫酸酯结合、葡萄糖醛酸结合等Ⅰ相及Ⅱ相代谢过程代谢为相应的产物。Objective:To study the metabolites of 5 flavonoids from the flowers of Trollius chinensis in vivo.Methods:Five typical flavonoid compounds from the flowers of T.chinensis including orientin,2″-O-β-L-galactopyranosylorientin,vitexin,2″-O-(2-methylbutanoyl)vitexin and isoquercetin were administrated intravenously and the rat plasma samples were obtained 10 min after administration.The high performance liquid chromatography-tandem mass spectrometry(HPLC-MS/MS)was used to investigate the metabolites in rat plasma.Results:A total of 20 metabolites had been identified and 5 of them were from orientin,2 of them were from 2″-O-β-L-galactopyranosylorientin,3 of them were from vitexin,5 of them were from 2″-O-(2-methylbutanoyl)vitexin and 5 of them were from isoquercetin,respectively.The structures of 20 metabolites and the metabolic pathways of 5 flavonoids were speculated on the basis of pseudo-molecular ion peaks and secondary fragments.Conclusion:Five compounds could be metabolized into the corresponding metabolites through phase I andⅡreactions involving oxidation,methylation,sulfating,glucuronidation,and so on.
