摘 要:目的:通过中心组合设计法(CCD)优化燥湿洗剂的提取工艺,以色谱法测定燥湿洗剂中黄芩苷的含量,提升其质量标准。方法:以黄芩中黄芩苷的含量为评价指标,通过响应面Plackett-Burman设计和CCD试验考察料液比、提取时间和提取次数3个主要影响因素对燥湿洗剂提取效果的影响,并对结果进行响应面分析,优化提取工艺。对燥湿洗剂中黄芩主要成分黄芩苷进行方法学考察,以验证CCD试验结果。结果:CCD优化得燥湿洗剂最佳提取工艺为料液比1∶10,提取时间60 min,提取数2次,黄芩苷的量可达1.1836 mg·mL^(-1),与模型预测值1.2042 mg·mL^(-1)相近。结论:综合运用响应面CCD联合色谱法优化燥湿洗剂的提取工艺,且优化所得工艺稳定、可行,优化的工艺参数可为其工业化生产提供参考。Objective:The quality standard of the Zaoshi Lotion was improved by optimization of the extraction process of lotion with CCD method,and determining the content of baicalin by chromatographic method.Methods:The content of baicalin,as the evaluation index,was detected in Plackett-Burman design and comined central composite of response surface methodology.Three pivotal influencing factors,such as the solid-to-liquid ratio,the time of extraction and extraction times,were screened and analyzed by response surface methodology.Results:The optimum Zaoshi Lotion extraction conditions were as follows:the solid-to-liquid ratio was 1∶10,the time of extraction was 60 min,extraction times were 2 times,the concentration of baicalin reached 1.1836 mg·mL^(-1),closely to the model predicted value of 1.2042 mg·mL^(-1).Conclusion:The extraction technology of Zaoshi Lotion has been optimized by combining CCD with chromatography method.The optimized extraction technology is stable and feasible and the optimized parameters could provide scientific basis for its further industrial production.
