摘  要:目的观察丹参饮预处理对缺血/再灌注损伤大鼠心肌细胞的保护作用及对血浆氧化三甲胺(TMAO)水平的影响。方法将50只SD大鼠随机分为假手术组、模型组及丹参饮低剂量组、中剂量组、高剂量组,各10只。采用前降支结扎法制备大鼠心肌缺血/再灌注损伤模型,观察心电图表现、心肌酶标志物、N-末端脑钠肽前体(NT-proBNP)、血浆TMAO浓度;苏木精-伊红(HE)染色观察心肌病理学改变。结果与模型组比较,丹参饮预处理组能降低大鼠血清肌酸激酶同工酶(CK-MB)、乳酸脱氢酶(LDH)与NT-proBNP水平(P<0.01),改善心肌病理学形态,且随丹参饮浓度增加改善作用更显著。与假手术组比较,模型组与丹参饮预处理组血浆TMAO浓度增高(P<0.05);丹参饮预处理组与模型组血浆TMAO浓度比较,差异无统计学意义(P>0.05)。结论丹参饮预处理可降低缺血/再灌注损伤大鼠血清心肌酶学水平,减轻心肌细胞缺血/再灌注损伤,保护心功能,且存在剂量关系,但丹参饮预处理未降低缺血/再灌注损伤模型大鼠血浆TMAO水平。Objective To investigate the protective effects of Danshen Decoction preconditioning on cardiomyocytes in rats with ischemia/reperfusion injury and its effect on trimethylamine N-oxide(TMAO).Methods Fifty SD rats were randomly divided into the sham group,model group,low-dose Danshen Decoction group,medium-dose Danshen Decoction group,and high-dose Danshen Decoction group,with 10 rats in each group.The rat model of myocardial ischemia/reperfusion injury was established by anterior descending branch ligation.Electrocardiogram,myocardial enzyme markers,N-terminal brain natriuretic peptide precursor(NT-proBNP),plasma TMAO concentration were compared between two groups and the changes of myocardial pathology were observed by hematoxylin-eosin(HE)staining.Results Compared with the model group,the levels of creatine kinase isoenzyme(CK-MB),lactate dehydrogenase(LDH),and NT-proBNP in the Danshen Decoction preconditioning group significantly decreased(P<0.01),the myocardial pathological morphology was improved,and the improvement degree increased with increase of Danshen decoction concentration.Compared with the sham group,the level of TMAO in the model group and Danshen Decoction preconditioning group was significantly increased(P<0.05).There was no significant difference in plasma TMAO between the model group and Danshen Decoction preconditioning group(P>0.05).Conclusion Danshen Decoction preconditioning could decrease the level of serum myocardial enzymology in rats with ischemia/reperfusion injury,reduce the myocardial ischemia/reperfusion injury,and protect cardiac function with depending dosage.Danshen Decoction pretreatment could not decrease the level of TMAO in rats with ischemia/reperfusion injury.
