猪精液中PCV-2 BJQX2020株基因图谱的构建与进化分析

摘　　要：为了了解猪精液中猪圆环病毒2(Porcine circovirus 2,PCV-2)基因组的分子特征,试验采用PCR技术对贵州省毕节市黔西县猪场种公猪精液中提取的核酸进行扩增克隆,通过预测开放阅读框(ORF)和酶切位点,以Winplas 2.7软件构建PCV-2 BJQX2020株的基因图谱,应用DNAStar中的MegAlign软件分析PCV-2 BJQX2020株基因组核苷酸序列和ORF2推导的氨基酸序列。结果表明:PCV-2 BJQX2020株的全基因组大小约为1 767 bp,为PCV-2b型;与国内外参考株全基因组核苷酸序列同源性为94.2%~100%,其中与国产疫苗株ZJ、SH、LG、DBN-SX07-2的同源性为95.6%~98.8%;ORF2氨基酸序列与国内外参考株的同源性为98.3%~100%;在1 069,1 071,1 138 bp碱基处的突变频率为25%。说明精液中PCV-2基因较为保守,变异不大,与大多数有临床症状表现的母猪、仔猪中PCV-2基因具有很高的同源性。In order to understand the molecular characteristics of Porcine circovirus 2(PCV-2) genome in swine semen, PCR technology was used to amplify and clone the nucleic acid extracted from boar semen of a swine farm in Qianxi County Bijie City Guizhou Province. Winplas 2.7 software was used to construct the gene map of PCV-2 BJQX2020 strain by predicting open reading frame(ORF) and restriction site prediction. The genome nucleotide sequence and ORF2-derived amino acid sequence of PCV-2 BJQX2020 strain were analyzed by MegAlign in DNAStar software. The results showed that the whole genome size of PCV-2 BJQX2020 strain was about 1 767 bp, and it was PCV-2 b type. The nucleotide sequence homology was 94.2%-100% with reference strains at home and abroad, 95.6%-98.8% with domestic vaccine strains ZJ, SH, LG, DBN-SX07-2. The amino acid sequence of ORF2 had 98.3%-100% homology with reference strains at home and abroad. The mutation frequency at 1 069, 1 071, 1 138 bp bases was 25%. It indicated that the PCV-2 gene in semen was relatively conservative with little variation, had high homology with the PCV-2 gene in most sows and piglets with clinical symptoms.
