摘  要:目的:对比研究三七细粉与破壁粉对高脂血症模型大鼠的作用,并初步探讨其作用机制。方法:采用高脂复合饮酒诱导高脂血症大鼠模型,造模成功后灌胃给予三七细粉0.15、0.3、0.6 g/kg以及破壁粉0.15、0.3、0.6 g/kg,每天给药1次,连续20 w。给药期间,测定大鼠血清总胆固醇(TC)、三酰甘油(TG)、高密度脂蛋白胆固醇(HDL-c)、低密度脂蛋白胆固醇(LDL-c)的含量,以及丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)活性;苏木素-伊红(HE)和油红O染色观察肝脏病变及脂质沉积情况;酶联免疫吸附法(ELISA)测定血清中游离脂肪酸(NEFA)、载脂蛋白A(Apo-A)、载脂蛋白B(Apo-B)以及肝脏中TC、TG、TBA和粪便TC、TBA水平;采用RT-PCR检测肝脏中肝X受体α亚型(lxrα)、三磷酸结合盒转运蛋白A1(abca1)、三磷酸结合盒转运体G1(abcg1)的mRNA表达。结果:与正常对照组比较,模型对照组大鼠血清TC、LDL-c含量、AST、ALT活力及肝脏TC、TBA水平明显升高(P<0.05或P<0.01);肝组织病变及脂质沉积严重,血清HDL-c、Apo-A水平明显降低及肝脏abca1、abcg1 mRNA表达明显下调(P<0.05或P<0.01);与模型对照组比较,三七细粉和破壁粉均能降低高脂血症模型大鼠血清中TC、TG、LDL-c及肝脏中TC、TBA水平,升高Apo-A和粪便TBA及尾部微循环水平(P<0.05或P<0.01);减轻大鼠肝脏脂肪空泡、炎性细胞浸润及脂质沉积(P<0.05或P<0.01);能上调肝脏abca1和abcg1 mRNA表达,下调lxrα mRNA表达(P<0.05或P<0.01),且破壁粉的作用强于细粉。结论:三七破壁粉调节血脂的作用优于细粉,主要表现在改善肝功能及肝脏脂质沉积上,其作用机制可能与调节lxrα、abca1、abcg1 mRNA表达而促进胆固醇流出有关。Objective: To compare the effects of Panax notoginseng powder and cell wall-broken powder on hyperlipidemia model rats, and to preliminarily explore its mechanism. Methods: The hyperlipidemia rat model was induced by high-fat diet combined with alcohol. After the model was successfully established, 0.15,0.3,0.6 g/kg Panax notoginseng powder and 0.15,0.3,0.6 g/kg cell wall-broken powder were administered once a day for 20 consecutive weeks. During the administration period, the contents of rat serum total cholesterol(TC),triglyceride(TG),high-density lipoprotein cholesterol(HDL-c),low-density lipoprotein cholesterol(LDL-c) and the activities of alanine aminotransferase(ALT) and aspartate aminotransferase(AST) were determined. Hematoxylin-eosin(HE) and oil red O staining were used to observe liver lesions and lipid deposition. Enzyme-linked immunosorbent assay(ELISA) was used to determine the serum levels of Free fatty acid(NEFA),apolipoprotein A(Apo-A) and apolipoprotein B(Apo-B),the liver levels of TC,TG and TBA, and stool TC,TBA levels. RT-PCR was used to detect the mRNA expressions of body alpha subtype(LXRα) in liver X receptor, ATP-binding cassette transporter A1(ABCA1),and triphosphate-binding cassette transporter G1(ABCG1). Results: Compared with the control group, the serum levels of TC,LDL-c, the activities of AST,ALT,and the liver levels of TC and TBA were significantly increased in the model group(P<0.05 or P<0.01). Liver tissue lesions and the lipid deposition were observed, the serum levels of HDL-c and Apo-A were decreased, the expressions of ABCA1 and ABCG1 mRNA in liver tissues were decreased(P<0.05 or P<0.01). Compared with the model group, the serum levels of TC,TG,LDL-c and liver levels of TC and TBA levels of hyperlipidemia model rats were reduced in Panax notoginseng powder and cell wall-broken powder groups, the Apo-A,fecal TBA and tail microcirculation levels were increase(P<0.05 or P<0.01).
