摘　　要：目的为了进一步研究糖尿病心肌病发病机制及其药物的作用机制。方法引进膜内皮蛋白(endoglin)敲除小鼠10雌6雄,进行扩繁,目的是获得足够数量的endoglin敲除小鼠的动物模型。本实验采用雌雄2∶1的比例合笼扩繁,从仔鼠尾尖中提取基因组DNA,进行PCR鉴定。结果成功获得两种基因型的子代小鼠,分别是endoglin敲除杂合子(endoglin^(+/-))和同窝野生型(WT)(纯合子在胚胎发育11d死亡)。结论正确的饲养繁殖管理及基因型鉴定方法可成功获得数量较多的雄性endoglin^(+/-)小鼠和同窝雄性WT(糖尿病模型常用雄性动物造模)。Objective In order to further study the pathogenesis of diabetic cardiomyopathy and the mechanism of drug action.Methods 10 females and 6 males of Endoglin knockout(Eng-iKOu)mice were introduced to expand propagation to obtain sufficient numbers of Eng-iKOu mice.In this experiment,the ratio of male-to-female cage propagation was 2∶1,and genomic DNA was extracted from the tail tips of young mice for genotyping by PCR.Results The successful offspring mice of two genotypes were Endoglin knockout heterozygote(Endoglin^(+/-))and homologous wild type(WT)(homozygote died at 11 days after embryonic development).Conclusion The results showed that a large number of male endoglin^(+/-)mice and littering male WT(diabetes model is often modelled by male animals)could be successfully obtained by proper breeding and breeding management and genotype identification.
