摘  要:目的:探讨P2X3受体抑制剂磷酸吡哆醛-6-偶氮苯-2,4-二磺酸(PPADS)治疗前列腺炎导致大鼠膀胱过度活动症的可行性。方法:24只SD大鼠随机数字法分为3组,PPADS组、生理盐水组、对照组。前列腺内注射1%角叉菜胶溶液建立前列腺炎大鼠模型。PPADS组采用PPADS膀胱灌注治疗,生理盐水组采用生理盐水膀胱灌注治疗,对照组不做任何处理。采用免疫印迹技术检测治疗前后大鼠膀胱组织中P2X3受体蛋白表达。尿动力学检测治疗前后大鼠膀胱排尿功能改变。组间比较采用t检验。结果:前列腺炎大鼠排尿频率为(23.35±2.47)次/h,不稳定收缩频率为(21.80±2.80)次/h,对照组排尿和不稳定收缩频率分别为(12.10±0.49)次/h和(1.60±0.70)次/h,前列腺炎大鼠排尿及不稳定收缩频率均高于对照组,差异有统计学意义(t=16.970,P<0.05)。PPADS灌注治疗后前列腺炎大鼠排尿频率为(12.97±1.02)次/h,不稳定收缩频率为(2.75±1.00)次/h,灌注治疗前后差异有统计学意义(t=17.910,P<0.05)。生理盐水组灌注治疗后排尿和不稳定收缩频率分别为(19.60±2.13)次/h和(18.50±2.50)次/h,灌注治疗前后差异无统计学意义(t=2.172、2.699,P>0.05)。前列腺炎大鼠膀胱组织内P2X3的表达量高于对照组。PPADS灌注治疗后前列腺炎大鼠膀胱组织内P2X3的表达量减少。结论:前列腺内注射角叉菜胶溶液可成功建立前列腺炎大鼠模型。前列腺炎可引起膀胱组织P2X3受体的表达上调。PPADS治疗可降低膀胱组织P2X3受体表达,抑制前列腺炎导致的膀胱过度活动。Objective To explore the feasibility of the P2X3 receptor inhibitor pyridoxalphosphate-6-azo benzene-2,4-disulfonic acid(PPADS)to treat prostatitis-induced overactive bladder in rats.Methods Totally,24 Sprague Dawley rats were randomly divided into three groups:PPADS group,saline group,and control group.A rat model of prostatitis was established by injecting 1%carrageenan solution into the prostate.The PPADS group was given bladder perfusion of PPADS,the saline group was given saline bladder perfusion,and the control group did not do any treatment.Western blotting was used to detect the expression of P2X3 receptor protein in rat bladder tissue before and after treatment.Urodynamic tests were used to test the changes of bladder voiding function in rats before and after treatment.Normally distributed measurement data were expressed as mean±standard deviation(±s)and analyzed by SPSS 22.0 statistical software,and comparison between groups was performed by t test.Results The urination frequency and the frequency of unstable contractions of prostatitis rats were(23.35±2.47)times/h and(21.80±2.80)times/h,significantly higher than in the control group[(12.10±0.49)times/h and(1.60±0.70)times/h respectively(t=16.970,P<0.05).After PPADS perfusion,the frequency of urination was(12.97±1.02)times/h,and the frequency of unstable contractions was(2.75±1.00)times/h,which were significantly different from those before perfusion treatment(t=17.910,P<0.05).In the saline group,the frequency of urination and unstable contraction after perfusion was(19.60±2.13)times/h and(18.50±2.50)times/h,respectively.There was no significant difference before and after the perfusion treatment(t=2.172,2.699,P>0.05).The expression of P2X3 in the bladder tissue of the prostatitis rats was higher than that of the control group.After PPADS perfusion treatment,the expression of P2X3 decreased in the bladder tissue of the prostatitis rats.Conclusion Injecting carrageenan solution into the prostate can successfully establish a rat model of prostat
