摘 要:目的:观察银杏叶提取物对急性心肌梗死大鼠凋亡水平的影响。方法:60只SD大鼠随机数字表法分为对照组,模型组和实验组,其中模型组和实验组采用结扎左冠状动脉前降支的方法复制AMI大鼠模型,对照组大鼠不结扎处理。建模成功后,实验组大鼠给予每天150 mg/kg银杏叶提取物,对照组和模型组给予等体积生理盐水。连续给药6 d后,采用超声影像检测3组大鼠心肌功能;采用苏木精-伊红(HE)染色法观察心肌组织病理学变化及进行损伤评分,采用脱氧核糖核苷酸末端转移酶介导的缺口末端标记法(TUNEL)分析心肌细胞凋亡指数;采用蛋白质印迹法(Western blot)分析3组大鼠心肌组织凋亡相关蛋白表达水平。计量资料分析采用单因素方差分析。结果:实验组大鼠舒张末期左室内径(LVEDD)和收缩末期左室内径(LVESD)[(7.28±0.30)、(5.00±0.28)mm]低于模型组[(8.01±0.36)、(7.15±0.34)mm],而射血分数(EF)水平[(58.41±5.09)%]高于模型组[(43.27±5.69)%],差异有统计学意义(t=2.781、3.017、3.001,P<0.05)。实验组大鼠心肌损伤评分[(4.02±0.57)分]显著低于模型组[(6.75±0.79)分],差异有统计学意义(t=3.793,P<0.05)。实验组大鼠心肌组织凋亡指数[(15.73±3.25)%]显著低于模型组[(33.97±3.96)%],差异有统计学意义(t=4.116,P<0.05)。实验组大鼠B细胞淋巴瘤/白血病-2相关X蛋白(bax)和半胱氨酰天冬氨酸特异性蛋白酶(Caspase)-3表达水平(1.31±0.18、2.09±0.19)低于模型组(1.98±0.21、3.15±0.28),差异有统计学意义(t=3.011、3.158,P<0.05)。结论:银杏叶提取物可显著下调凋亡相关蛋白表达,抑制心肌组织细胞凋亡,进而保护急性心肌梗死大鼠心脏功能。Objective To investigate the effect of Ginkgo biloba extract on apoptosis in rats with acute myocardial infarction.Methods Totally,60 Sprague Dawley rats were randomly divided into sham operation group,model group and experimental group by the random number table.The AMI rat model was established by ligating the left anterior descending coronary artery in the model group and experimental group,and the left anterior descending coronary artery in the sham operation group was not ligated.After successful modeling,rats in the experimental group were given 150 mg/kg Ginkgo biloba extract,and those in the sham operation group and model group were given the same volume of normal saline.After 6 days of continuous administration,the myocardial function of the three groups was detected by ultrasonic imaging.The myocardial histopathological changes and injury scores were observed by hematoxylin eosin staining(HE).The apoptosis index of myocardial tissue was analyzed by terminal deoxynucleotidyl transferase mediated nick end labeling(TUNEL).The expression levels of apoptosis related proteins in myocardial tissue of the three groups were analyzed by Western blotting.SPSS 17.0 software and one-way ANOVA were used to analyze the data.Results The left ventricular end diastolic diameter(LVEDD)and left ventricular end-systolic dimension(LVESD)in the experimental group[(7.28±0.30)and(5.00±0.28)mm]were lower than those in the model group[(8.01±0.36)and(7.15±0.34)mm],and ejection fraction(EF)level[(58.41±5.09)%]was higher than that in the model group[(43.27±5.69)%].The difference was statistically significant(t=2.781,3.017,3.001,P<0.05).The myocardial injury score in the experimental group[(4.02±0.57)points]was significantly lower than that in the model group[(6.75±0.79)points(t=3.793,P<0.05).The myocardial apoptosis index in the experimental group[(15.73±3.25)%]was significantly lower than that in the model group[(33.97±3.96)%,t=4.116,P<0.05].The expression levels of B cell lymphoma/leukemia-2 associated X protein(bax)and
