摘 要:目的测定补血益母丸的HPLC指纹图谱,并测定制剂中益母草碱、毛蕊异黄酮葡萄糖苷、芦丁、阿魏酸、藁本内酯等5种指标性成分的含量。方法采用Symmetry Shield TM RP18色谱柱(150 mm×3.9 mm,5μm),进行指标性成分含量测定方法的建立与确证;测定不同批次补血益母丸中5种指标性成分的含量,同时建立补血益母丸的HPLC指纹图谱,应用中药色谱指纹图谱相似度评价系统软件对10个批次样品进行相似度分析。结果10批补血益母丸样品中共确认了36个共有峰,指认出5个指纹峰,依次为益母草碱、毛蕊异黄酮葡萄糖苷、芦丁、阿魏酸、藁本内酯(对应1、2、3、4、5号峰),10批补血益母丸制剂与对照图谱的相似度均大于0.96。益母草碱、毛蕊异黄酮葡萄糖苷、芦丁、阿魏酸、藁本内酯等5种成分的线性关系良好(r≥0.9999),含量分别为0.1285~0.1689、0.1536~0.2705、0.1079~0.3011、0.1914~0.2809、1.8122~3.3256 mg·g^(-1),其中,以藁本内酯的含量较高。结论所用方法准确可靠、稳定可控,适用于补血益母丸的质量控制。OBJECTIVE To determine HPLC fingerprint of 10 batches of Buxue Yimu pills(BYP),and the major five compounds of BYP.METHODS HPLC analysis was performed with a C18 column(150 mm×3.9 mm,5μm).The methodology for determining the content of 5 representative components was validated.Then the content of the 5 representative components in 10 batches of BYP were detected.The similarity of chemical profile was evaluated by the similarity evaluation system of Traditional Chinese Medicine.RESULTS The HPLC fingerprint of 10 batches of BYP was established and 36 common fingerprint peaks were identified,and the similarity of 10 batches of BYP was greater than 0.96.The linearity relationships of leonurine,calycosin-7-O-glucoside,rutin,ferulic acid and ligustilide were good(r≥0.9999).The content of leonurine,calycosin-7-O-glucoside,rutin,ferulic acid and ligustilide in 10 batches of BYP were 0.1285-0.1689,0.1536-0.2705,0.1079-0.3011,0.1914-0.2809 and 1.8122-3.3256 mg·g^(-1),respectively.Among all the detected components,ligustilide was accounted for the highest content.CONCLUSION The method is accurate,stable,and suitable for the quality control of BYP.
