摘  要:目的探讨滋膵通脉饮对糖尿病大鼠血糖、胰岛素抵抗和骨骼肌磷脂酰肌醇-3激酶(PI3K)信号通路的影响。方法45只清洁级SD大鼠,随机数字表法抽取10只作为空白对照组,剩余35只采用高脂高糖饲料喂养及腹腔注射链脲佐菌素的方法构建2型糖尿病大鼠模型,将造模成功的糖尿病大鼠随机分为3组:模型组、滋膵通脉饮组和罗格列酮组,每组10只。于实验第7周开始灌胃,滋膵通脉饮组灌服滋膵通脉饮,生药量为17.6 g/kg;罗格列酮组灌服罗格列酮混悬液0.42 mg/(kg·d);空白对照组和模型组均灌服同等容积的蒸馏水,每日1次,共用药4周。4周后称取大鼠体重,测大鼠血糖、胰岛素,计算胰岛素抵抗指数,半定量PCR技术检测大鼠骨骼肌PI3K、GLUT4 mRNA表达。结果与空白对照组比较,模型组造模后大鼠体重升高,药物干预后降低(P<0.05);药物干预后,与模型组比较,滋膵通脉饮组和罗格列酮组体重明显上升(P<0.05)。与空白对照组比较,药物干预前模型组血糖升高(P<0.05)。药物干预后,滋膵通脉饮组和罗格列酮组血糖较模型组降低(P<0.05)。与空白对照组比较,模型组胰岛素和胰岛素抵抗指数升高,PI3K、GLUT4 mRNA表达降低(P<0.05);与模型组比较,滋膵通脉饮组和罗格列酮组胰岛素和胰岛素抵抗指数明显降低,PI3K、GLUT4 mRNA表达上升(P<0.05)。结论滋膵通脉饮降低糖尿病大鼠血糖、改善胰岛素抵抗的同时还能激活PI3K介导的信号通路,这可能是其降低血糖、改善胰岛素抵抗的主要机制之一。Objective To explore the effects of Zicui Tongmai Decoction(ZCTMD)on blood glucose,insulin resistance and phosphatidylinositol 3-kinase(PI3K)signaling pathway in skeletal muscle of diabetic rats.Methods Ten of the 45 SD rats(clean grade)were selected as control group using random number table,the remaining 35 rats were fed with high-fat and high sugar diet,combined with intraperitoneal injection of streptozotocin to establish the model of type 2 diabetes.The successfully established diabetic rats were randomly divided into three groups:model group,ZCTMD group and rosiglitazone group,with 10 rats in each group.Rats were administered through gastric gavage since the 7th week,once a day for 4 weeks.ZCTMD group was administered with ZCTMD decoction 17.6 g/kg(equivalent to the crude herb)intragastrically,rosiglitazone group was rosiglitazone suspension,0.42 mg/(kg·d).The control group and model group were given distilled water of the same volume.After 4 weeks,the rats were weighed,blood glucose and insulin were measured,insulin resistance index was calculated,and the expression of PI3K and GLUT4 mRNA in skeletal muscle was detected by semi-quantitative PCR.Results Compared with the control group,the body weight of rats in the model group was increased after modelling,but decreased after drug intervention(P<0.05).The body weight of rats in the ZCTMD group and the rosiglitazone group was increased significantly while comparing to the model group(P<0.05).The blood glucose of the model group was increased before drug intervention compared with control group(P<0.05).After drug intervention,the blood glucose of ZCTMD group and rosiglitazone group was significantly lower compared with the model group(P<0.05).The insulin and insulin resistance index in the model group were increased significantly,and the expression of PI3K and GLUT4 mRNA were decreased compared with the control group(P<0.05).Compared with the model group,the insulin and insulin resistance index in the ZCTMD group and rosiglitazone group were decreased,the ex
