摘 要:目的建立血液样本中氯苯的顶空-气相色谱测定方法。方法将大鼠血样直接置于顶空瓶中,采用顶空气相色谱法,Agilent DB-1(30.00 m×0.32 mm×3.00μm)毛细管色谱柱,优化顶空条件,以外标法定量。结果该方法在质量浓度为10-2 000μg/L时线性关系良好,相关系数为0.999 8;最低检出浓度为10μg/L。日内和日间精密度<15%,准确度在-5.3%--2.6%之间,回收率为88.2%-97.1%。大鼠经口灌胃200 mg/kg的氯苯后,其体内主要毒代动力学参数:半衰期(t1/2)为(2.804±0.496)h,曲线下面积[AUC(0-t)]为(66 225.3±11 926.8)μg/(L·h),平均驻留时间[MRT(0-t)]为(4.314±0.448)h,表观分布容积(Vz/F)为(12.44±2.71)L/kg,清除率(CLz/F)为(3.094±0.569)L/(h·kg)。结论该方法高效、灵敏、特异性强,准确度、精密度好,可用于氯苯的毒代动力学研究、职业暴露监测和中毒分析。Objective To develop a method of determination of monochlorobenzene in blood samples by headspace gas chromatography. Methods The blood sample was placed directly in the headspace bottle,monochlorobenzene was detected by headspace gas chromatography with Agilent DB-1(30.00 m×0.32 mm×3.00 μm) capillary column,the headspace conditions were optimized,and the external standard method was used for quantitative analysis. Results The standard curves were linear over the concentration range of 10-2 000 μg/L,the correlation coefficient was 0.999 8,and the minimum detectable concentration was10 μg/L. The intra-and inter-day precision was 15%,the accuracy was-5.3%--2.6%,and the recovery was ranged from 88.2% to97.1%. After oral administration of 200 mg/kg monochlorobenzene in rats,main toxicokinetic parameters were as follows:half-life period(t1/2) was(2.804±0.496) h,area under curve [AUC(0-t)] was(66 225.3±11 926.8)μg/(L·h),average dwell time(MRT)(0-t)was(4.314±0.448)h,apparent distribution volume(Vz/F) was(12.44±2.71)L/kg,and clearance rate(CLz/F) was(3.094±0.569)L/(h·kg).Conclusion The method is highly efficient and sensitive,with high specificity,and good accuracy and precision,which is suitable to toxicokinetic study,occupational exposure monitoring and poisoning analysis of monochlorobenzene.
