十味降糖散对糖尿病大鼠肝脏、脂肪AMPKα和肾CTGF因子表达的调节

摘　　要：为研究十味降糖散对糖尿病大鼠肝肾保护和脂代谢的机制,采用链脲佐菌素(STZ)建立大鼠糖尿病模型,设空白组、模型Ⅰ组(糖尿病模型)、模型Ⅱ组(糖尿病肾病模型)、治疗Ⅰ组和治疗Ⅱ组,免疫印迹法检测大鼠肝脏、睾丸脂肪垫中腺苷酸活化蛋白激酶(AMPKα)的表达情况,免疫组织化学检测大鼠肾脏结缔组织生长因子(CTGF)的表达情况。结果显示:与空白组比较,模型Ⅰ组肝脏AMPKα表达极显著降低(P<0.01),与模型Ⅰ组比较,治疗Ⅰ组肝脏AMPKα表达显著升高(P<0.05),空白组和治疗Ⅰ组肝脏AMPKα表达无差异(P>0.05);与空白组比较,治疗Ⅰ组和模型Ⅰ组睾丸脂肪垫AMPKα表达无差异(P>0.05);与空白组比较,模型Ⅱ组CTGF表达显著升高(P<0.05),与模型Ⅱ组相比,治疗Ⅱ组CTGF表达显著降低(P<0.05),空白组和治疗Ⅱ组CTGF表达无差异(P>0.05)。结果表明十味降糖散可以上调糖尿病大鼠肝脏AMPKα的表达,抑制肾脏CTGF的表达,对睾丸脂肪垫AMPKα的表达无影响,这可能与十味降糖散对肝脏的保护与肝脏AMPKα表达有关,对肾脏的保护作用与CTGF蛋白的表达有关,而改善脂代谢与睾丸脂肪垫AMPKα表达无关。To study the mechanism of Shiwei Jiangtang powder on liver and kidney protection and lipid metabolism in diabetic rats.A rat model of diabetes was established through streptozotocin(STZ)treatment.The rats were divided into blank group,model I group,model II group,treatment I group and treatment II group.The expression of AMPKαin rat liver and testicular fat pad was detected by immunoblotting,and the expression of CTGF in rat kidney was detected by immunohistochemical test.The results showed that compared with the blank group,the expression of AMPKαin the liver of model I group was significantly lower(P<0.01),and the expression of AMPKαin liver of treatment I group was significantly higher than that in model I group(P<0.05).There was no significant difference in the expression of AMPKαin the liver of blank group and treatment I group(P>0.05).Compared with the blank group,there was no significant difference in the AMPKαexpression of testicular fat pad in treatment I group and model I group(P>0.05).Compared with the blank group,the expression of CTGF in model II group increased significantly(P<0.05).Compared with modelⅡgroup,the expression of CTGF decreased significantly in treatment II group(P<0.05).While there was no difference in the CTGF expression between the blank group and treatment II group(P>0.05).The results showed that Shiwei Jiangtang powder could up-regulate the expression of AMPKαin liver and inhibit the expression of CTGF in kidney of diabetic rats,but had no effect on the expression of AMPKαin testicular fat pad.This indicates that the protective effect of Shiwei Jiangtang powder on liver may be related to the induction of AMPKαin liver,and that the protection of kidney may be relate to the expression of CTGF protein suppressed by Shiwei Jiangtang powder,but the improvement of lipid metabolism by Shiwei Jiangtang powder is not related to the modification of AMPKαexpression in the testicular fat pad.
