多指标综合评分结合正交试验法优选大鼠对乙酰氨基酚肝损伤模型

摘  要:采用正交试验方法优选大鼠对乙酰氨基酚(APAP)染毒致肝损伤模型最佳制作条件。选择对肝功能指标有影响的3个因素:染毒剂量、中毒途径和采血时间,设置3个水平,采用L_(9)(3^(4))正交表安排试验,以肝生化指标综合评分作为参考依据,优选大鼠肝损伤模型的最佳条件。结果显示,APAP以800 mg/(kg·bw)经腹腔注射雄性大鼠,28 h后可以得到最优化肝损伤模型。本方法可为肝损伤新药研发和机理研究提供可靠的模型。To optimize the preparation conditions of acetaminophen induced liver injury in rats by orthogonal test.Three factors affecting liver function were selected:(1)the dose of paracetamol,(2)the way of poisoning,(3)the time of blood collection,and each has selects three levels.Experiments were designed according to L_(9)(3^(4))orthogonal test table.Based on the liver biochemical indexes,the best conditions for making liver injury models in rats were optimized.The resutts showed that when male rats were injected intraperitoneally with paracetamol 800 mg/(kg·bw),after 28 hours,the optimal liver injury model was established.This method has good reproducibility,and can be used as a reliable model for studying liver injury and liver damage mechanism of drug development.
