摘  要:为揭示连云港基岩岸常见帽贝物种的丰富度,以细胞色素c氧化酶Ⅰ(COⅠ)基因条形码为主要工具,参考世界海洋物种目录数据库(World Register of Marine Species,WoRMS),对连云港连岛西侧分布的帽贝物种进行了识别。共识别4种帽贝,分别为:日本菊花螺(Siphonaria japonica)、笠贝(Lottia goshimai)、齿舌新笠贝(Nipponacmea radula)、圆锥拟帽贝(Patelloida conulus)。其中,利用COⅠ条形码准确识别出3个物种:日本菊花螺、笠贝、齿舌新笠贝(经与NCBI数据库的序列比对,相似度均≥99%);借助16S rRNA条形码可进一步识别出圆锥拟帽贝。日本菊花螺与笠贝为该地区首次报道种。研究结果为连云港地区帽贝物种多样性研究提供了重要的参考资料。Limpets living at the coast are of great value in both academic and applied fields.However,few studies on the status of these species at the coast of Lianyungang are available.To investigate the richness of limpets in the area,this study identified limpets mainly by using COⅠ(cytochrome c oxidaseⅠ)barcoding and referring to pictures from the database of the World Register of Marine Species(WoRMS)after sample collection in four seasons.Results showed that four species of limpets were identified in this study(Siphonaria japonica,Lottia goshimai,Nipponacmea radula,Patelloida conulus).Of them,Siphonaria japonica(sample ID X-W-20)was the most common limpet with high proportion in a certain sampling season(51.4%in spring,49.2%in summer,49.2%in autumn,64.3%in winter),indicating the potential usage of S.japonica to be served as the suitable biomarker in researching coastal environmental changes.Totally,three species can be accurately identified based on COⅠbarcoding:S.japonica,L.goshimai and N.radula(compared with sequences in NCBI database,the identity was≥99%).On the other hand,in the phylogenetic analysis,samples X-W-7 and X-W-18 were mixing in an ambiguous clade,which included P.conulus and N.schrenckii.The identification of X-W-7 and X-W-18 required other tools such as 16S rRNA barcoding(results from our unpublished data showed that the first three sequences were from P.conulus by blasting,and the identity was≥96%).Species of S.japonica and L.goshimai(firstly named by Japanese researchers in 2017)were firstly reported in Lianyungang.This study has improved the diversity of limpets for this region.According to this study,we expect that there should be more than four limpet species on this rocky intertidal shore,and this will be further verified in the future.
