猪血凝性脑脊髓炎病毒感染对小鼠脑内星形胶质细胞的影响

摘　　要：为阐明猪血凝性脑脊髓炎病毒(porcine hemagglutinating encephalomyelitis virus, PHEV)感染后小鼠脑内星形胶质细胞的活化情况,本研究通过滴鼻途径将PHEV接种BALB/c小鼠,分别运用qRT-PCR、Western blot和间接免疫荧光方法检测感染后不同时间点小鼠脑组织内星形胶质细胞特异性标志物GFAP的表达情况。结果表明,与正常对照组小鼠相比,接种PHEV后3 d即可检测到GFAP表达水平显著升高,并且GFAP的表达量随着感染时间延长逐渐增加。间接免疫荧光结果发现星形胶质细胞数量增加,分布集中,同时细胞体积增大,突起变多,证明星形胶质细胞在PHEV感染的小鼠脑内发生了活化。上述结果可为后期深入开展PHEV所致神经炎性损伤的机制研究奠定基础。To clarify whether astrocytes are activated in the central nervous system of mice with PHEV infection, 3-week-old BALB/c mice were inoculated with PHEV or PBS by intranasal route.GFAP was examined by using qRT-PCR,Western blot and indirect immunofluorescence(IF) at different time points after PHEV infection.The results showed that the expression level of GFAP of mice infected with PHEV was significantly increased on the 3 rd day after inoculation with PHEV compared with control mice, and the expression level of GFAP increased gradually over time.The results of IF further confirmed the activation of astrocytes in PHEV infected mice, which was characterized by increased cell number and concentrated distribution, as well as cell hypertrophy and proliferation.These results indicated that astrocytes are activated in the brain of mice infected with PHEV.This study lays a foundation for the study of neuroinflammatory injury caused by PHEV infection.
