摘  要:m6A甲基化修饰广泛存在于真核细胞和病毒RNA中,在病毒感染过程及宿主细胞抗病毒反应中扮演重要角色,然而m6A修饰在新城疫病毒复制过程中发挥的功能并不清楚。本研究发现新城疫病毒(NDV)感染后显著增加HD11细胞RNA的整体甲基化水平,抑制甲基转移酶METTL3的表达而促进甲基转移酶METTL14的表达。进一步研究发现,抑制m6A甲基化修饰显著促进NDV NP基因的mRNA转录及翻译,过表达甲基转移酶METTL3和METTL14明显抑制NDV的复制,表明m6A甲基化修饰在NDV复制过程中起负调控作用。Methylation modification of N6-methyladenosine(m6 A) are widely present in eukaryotic cells and viral RNA,it plays an important role in the process of virus infection and the antiviral response of host cells.However, the function of m6 A in the replication of Newcastle disease virus(NDV) is not clear.In this study, we found that NDV infection significantly increased the overall RNA methylation level of HD11 cells, inhibited the expression of methyltransferase METTL3 and promoted the expression of methyltransferase METTL14.Further studies showed that inhibition of m6 A methylation significantly promoted mRNA transcription and translation of the NDV NP,and overexpression of methyltransferases METTL3 and METTL14 significantly inhibited the replication of NDV,suggesting that m6 A methylation plays a negative regulatory role in NDV replication.
