山羊支原体山羊肺炎亚种在2种培养基上的生长特性比较

摘  要:为筛选出山羊传染性胸膜肺炎疫苗制备中抗原产量高且成本低的培养基,本试验将山羊支原体山羊肺炎亚种M1801株和M1601株接种于含20%马血清的改良Thiaucourt’s肉汤(MTB)和新研制的含5%马血清的培养基(HF3),在120 h内监测其pH值、颜色变化单位(color change units, CCU)和菌体蛋白浓度的变化,绘制相应曲线。结果表明,山羊支原体山羊肺炎亚种用HF3培养时pH值下降更慢,2个菌株在HF3培养基中的CCU计数均高于MTB培养基,且高滴度维持时间更长,菌体的蛋白浓度也略高于MTB培养基。结果提示山羊支原体山羊肺炎亚种在HF3培养基中产量更高,较MTB具有更好的效果,且血清浓度低,成本更低廉,为山羊传染性胸膜肺炎灭活疫苗的大批量生产奠定了基础。In order to screen the medium with high antigen yield and low cost in the preparation of contagious caprine pleuropneumonia vaccine, Mycoplasma capricolum subsp. Capripneumoniae(Mccp) M1801 and M1601 strains were inoculated in modified Thiaucourt’s broth(MTB) containing 20% horse serum and the newly developed medium(HF3) containing 5% horse serum.The pH,color change units(CCU) and bacterial protein concentration were monitored within 120 h, and the corresponding curve were drawn.The results showed that when Mccp was cultured in HF3 medium, the pH decreased more slowly.The CCU counts of the two strains in the HF3 medium were higher than that in MTB medium, and the high titer was maintained longer.The protein concentration was also slightly higher than that in MTB medium.The results suggested that Mccp had higher yield in HF3 medium, and the serum concentration and cost of HF3 medium were lower than MTB,which lays a foundation for mass production of contagious caprine pleuropneumonia inactivated vaccine.
