摘 要:从本实验室前期甲型流感病毒(inlfuenza A virus, IAV)感染的环状RNA(circular RNA,circRNA)组学数据中遴选出1个差异显著上调的circRNA hsa_circ_0070421,在此探讨其与IAV感染之间的关联以及对病毒复制的影响。首先运用荧光定量PCR等方法鉴定hsa_circ_0070421的成环,确证IAV感染与A549细胞内hsa_circ_0070421表达的关联;其次用Poly I:C、LPS、干扰素(interferon, IFN)等处理细胞以确定IAV感染所致hsa_circ_0070421表达上调的诱因;最后通过hsa_circ_0070421的过表达和敲低试验来明确其对IAV复制的影响。结果显示,宿主hsa_circ_0070421的表达上调与IAV感染复制呈显著正相关,poly I:C和IFN-β处理均可诱导A549细胞内hsa_circ_0070421的表达显著上调,并且hsa_circ_0070421具有抑制IAV复制的功能。结果表明,hsa_circ_0070421是宿主抗IAV天然免疫应答的产物,可能是Ⅰ型IFN抗IAV的手段之一。From the preliminary data on circular RNA(circRNA) expression profiling, hsa_circ_0070421 that was significantly up-regulated was selected, its roles in influenza A virus(IAV) infection and replication were further explored.Firstly, the existence of circular hsa_circ_0070421 was validated by RT-PCR,and the correlation between IAV infection and hsa_circ_0070421 expression in A549 cells was confirmed using qRT-PCR;Secondly, we determined the trigger of hsa_circ_0070421 upregulation in A549 cells treated with Poly I:C,LPS,interferon, etc.respectively;Finally, overexpression and knockdown of hsa_circ_0070421 were used to clarify its impact on IAV replication.The results showed that there was a positive correlation between hsa_circ_0070421 expression and IAV infection.Poly I:C and interferon treatment induced the expression of hsa_circ_0070421.Hsacirc0070421 inhibited IAV replication at mRNA,protein and progeny virion levels, respectively.In summary, our data suggested that hsa_circ_0070421,as a host response to IAV infections, plays an antiviral role against IAV and our study provides a new sight to understand IAV-host interaction.
