摘  要:通过Illumina MiSeq高通量测序技术,对花生根、茎、叶、花不同器官内生细菌16S rRNA的V5~V7可变区进行测序,研究花生内生细菌群落多样性。从花生不同器官内一共获得61396条有效序列和468个OTU(Operational Taxonomic Units)。物种分类显示花生内生细菌种类隶属于21个门、43个纲、107个目、177个科和284个属。根部内生细菌优势属为假单胞菌属(Pseudomonas)、慢生根瘤菌属(Bradyrhizobium)和魏斯氏菌属(Weissella),相对丰度分别为37.67%、25.13%和7.38%;茎部内生细菌优势属为假单胞菌属(Pseudomonas)和红球菌属(Rhodococcus),相对丰度分别为73.12%和8.88%;叶部内生细菌优势属为假单胞菌属(Pseudomonas)和红球菌属(Rhodococcus),相对丰度分别为78.65%和8.55%;花部内生细菌优势属为假单胞菌属(Pseudomonas),相对丰度为15.15%。PICRUSt功能预测表明,叶部内生细菌中参与调控各种代谢通路的物种丰度最高,茎部和根部次之,花部最低。以上研究结果表明花生内生细菌具有丰富的种群多样性,为今后开发利用花生内生细菌资源奠定了基础。In order to investigate the microbial diversity of endophytic bacterial community in different organs of peanut,the V5-V7 regions of endophytic bacterial 16S rRNA genes from peanut root,stem,leaf and flower were sequenced by Illumina MiSeq high-throughput sequencing technology.A total of 61,396 valid sequences and 468 OTUs were obtained from different organs of peanut.Species classification showed that endophytic bacteria in peanut belonged to 21 phyla,43 classes,107 orders,177 families and 284 genera.The predominant genera in root were Pseudomonas、Bradyrhizobium and Weissella with relative abundance of 37.67%,25.13%and 7.38%,respectively.The predominant genera in stem were Pseudomonas and Rhodococcus with relative abundance of 73.12%and 8.88%,respectively.The predominant genera in leaf were Pseudomonas and Rhodococcus with relative abundance of 78.65%and 8.55%,respectively.The predominant geneus in flower was Pseudomonas with relative abundance of 15.15%.PICRUSt function prediction analysis showed that the species richness of leaf endophytic bacteria involved in the regulation of various metabolic pathways was the highest,followed by stem and root,and the lowest was in flower.The results showed that endophytic bacteria in peanut had rich population diversity,which laid a foundation for the development and utilization of endophytic bacteria resources in peanut in the future.
