摘 要:目的:应用毛细管电泳技术测定天山堇菜中5种活性成分(白杨素、七叶内酯、伞形花内酯、山柰酚和槲皮素)的含量。方法:对影响毛细管电泳分离的主要参数进行优化,包括缓冲溶液的种类及浓度、添加剂的种类及浓度、分离电压和检测波长,得到最优分离条件。结果:最优缓冲条件为缓冲溶液为30 mmol/L硼砂溶液-20%乙腈,分离电压为12 kV,电泳时间为30 min,0.5 psi(3.45 kPa)压力进样5.0 s,紫外检测波长为198 nm。结果:本方法线性良好(R^(2)=0.9920~0.9990),方法重现性高(RSD<5.7%),回收率为98.2%~110.2%;天山堇菜中七叶内酯含量为20~50μg/g,伞形花内酯含量为30~70μg/g,山柰酚含量为220~250μg/g。结论:毛细管电泳法可以快速、方便测定天山堇菜中活性成分的含量,并为毛细管电泳技术在中草药中活性成分的测定提供了应用依据。OBJECTIVE:To determine the content of 5 active components(chrysin,esculin,umbelliferone,kaempferol and quercetin)in Viola tianshanica by capillary electrophoresis.Method:The main parameters affecting the separation of capillary electrophoresis were optimized,including the type and concentration of buffer solution,the type and concentration of additives,the separation voltage and the detection wavelength,and the optimal separation conditions were obtained.Results:The optimal buffer conditions were 30 mmol/L borax solution-20%acetonitrile,separation voltage of 12 kV,electrophoresis time of 30 min,0.5 psi pressure injection for 5.0 s,and UV detection wavelength of 198 nm.Results:The method has good linearity(R^(2)=0.9920~0.9990),high reproducibility of the method(RSD<5.7%),and the recovery rate is 98.2%~110.2%;the content of esculetin in Viola tianshanica is 20~50µg/g,umbelliferone content is 30~70µg/g,kaempferol content is 220~250µg/g.Conclusion:Capillary electrophoresis can quickly and conveniently determine the content of active ingredients in Viola tianshanica,and provides a basis for the application of capillary electrophoresis in the determination of active ingredients in Chinese herbal medicine.
