摘  要:双光子显微镜是结合了激光扫描共聚焦显微镜和双光子激发技术的一项新技术,具有探测能力强、光漂白小的特点,在活体动物脑血管研究中具有特殊优势。该文介绍了双光子显微镜的基本原理和组成,并从仪器开机、操作流程、注意事项等方面介绍了双光子显微镜的使用方法。详细阐述了双光子显微镜在脑血管研究中检测方法的建立和注意事项。具体包括观察脑血管立体分布和增值,量化血流速度,检测白细胞与内皮细胞的相互作用,构建脑血栓动物模型。建立了利用双光子显微镜检测活体动物脑血管平台,为脑疾病基础研究和创新药物开发提供了先进的技术手段。The two-photon microscope is a new technology that combines laser scanning confocal microscope and two-photon excitation technology. It has the characteristics of strong detection ability and low-photo bleaching with special advantages in the detection of cerebral vessels in live animals. This paper introduces the basic principle and composition of two-photon microscope, and the use of two-photon microscope from the aspects of instrument startup, operation process and precautions. The establishment and precautions of two-photon microscope detection methods in cerebrovascular research are elaborated in detail. Specifically, it includes studying the three-dimensional distribution and value-added of cerebral blood vessels, quantifying blood flow velocity, detecting leukocyte-endothelial cell interactions, and constructing animal models of cerebral thrombosis. A two-photon microscope platform to detect the cerebral vessels of living animals is established to provide advanced technical means for basic research of brain diseases and the development of innovative drugs.
