复合益生菌对K99引起的犊牛腹泻小肠结构及肠胶质细胞分泌亚硝基谷胱甘肽的影响

摘  要:本研究旨在探究复合益生菌对大肠杆菌K99引起的犊牛腹泻小肠病理变化及肠胶质细胞(EGC)分泌亚硝基谷胱甘肽(GSNO)的影响。犊牛随机分为三组:对照组犊牛灌服10mL磷酸缓冲盐溶液;益生菌组犊牛灌服K99菌液10mL及饲喂复合益生菌;感染组犊牛灌服大肠杆菌K9910mL,分别于7d、14d、21d取组织样用苏木精-伊红染色法染色观察,酶联免疫吸附法(ELISA)测定各组犊牛小肠GSNO含量。结果表明:与对照组相比,益生菌组各段小肠绒毛结构完整,无病理变化,感染组各段小肠肠绒毛断裂及水肿的较多;ELISA结果显示,十二指肠中GSNO含量各处理组间无显著差异(P>0.05);空肠中益生菌组GSNO含量最高,与对照组间无显著差异(P>0.05),感染组GSNO含量最低,且显著低于对照组和益生菌组(P<0.05);回肠中感染组GSNO含量极显著低于对照组和益生菌组(P<0.01),益生菌组与对照组间差异不显著(P>0.05)。试验表明K99可导致小肠组织病理学变化,降低GSNO的分泌;益生菌可保护犊牛小肠组织结构,提高GSNO的分泌。The purpose of this study was to investigate the effects of compound probiotics on intestinal pathology of diarrhea in calves caused by Escherichia coli K99 and the secretion of nitrosoglutathione(GSNO)caused by enteric glial cell(EGC).Calves were randomly divided into three groups.The control group was given 10ml phosphate buffer saline(PBS).Group 1 of calves was fed K99 bacteria solution and compound probiotics.And calves in test group 2 were given Escherichia coli K9910ml by gavage 1;Tissue samples were taken at 7,14 and 21 days for HE staining,and the GSNO content of calves in each group was determined by ELISA.The results showed that compared with the control group,the villus of each segment of the test group 1 were intact and had no pathological changes.There were more villus rupture and edema in each segment of the small intestine in the 2 groups.ELISA results showed that there was no significant difference in GSNO content in the duodenum(P>0.05);and in jejunal test group 1 has the highest GSNO content,while,in test group 2 was the lowest,which was significantly different from that in control group and test group 1(P<0.05),and there was no significant difference between test group 1 and control group(P>0.05);In the ileum compared with the control group,there were significantly differences between the two groups(P<0.01),there was no significant difference in test group 1(P>0.05).The results showed that K99 could lead to Histopathology of small intestine and decrease the secretion of GSNO.While probiotics can protect the small intestine structure of calves and improve the secretion of GSNO.
