摘  要:目的采用UPLC-MS/MS方法评估酒精干预百草枯急性中毒后的大鼠代谢组学变化。方法雄性SD大鼠60只,随机分为6组,提前酒精干预组、同时酒精干预组、百草枯组、酒精组、对照组、长期酒精干预组。提前酒精干预组先给予酒精,后给予百草枯;酒精干预组同时给予酒精与百草枯;百草枯组给予百草枯;酒精组给予酒精;对照组给予生理盐水;长期酒精干预组,先给予酒精4周后,再给予百草枯。在给予12 h后采血2 ml,离心后取血清,处理后,运用UPLC-MS/MS分析。结果与对照组相比,百草枯中毒后,乳酸、苏氨酸、丙氨酸和腺嘌呤升高(P<0.05),谷氨酰胺、丙酮酸、色氨酸和苯丙氨酸降低(P<0.05);提前酒精干预组和同时酒精干预组,乳酸都变得更高(P<0.01),长期酒精干预后的乳酸降低一些(P<0.05);酒精干预百草枯后的3组,丙酮酸、色氨酸浓度都有回升,与对照组差异无统计学意义(P>0.05)。结论相比提前酒精干预组和同时酒精干预组,长期饮酒后干预引起的代谢物变化更小,但是其机制需要进一步研究。Objective To evaluate the metabolomics changes of acute paraquat after alcohol intervention using ultra performance liquid chromatography-tandem mass spectrometry(UPLC-MS/MS).Methods Sixty male SD rats were randomly divided into six groups:early alcohol intervention group,simultaneous alcohol intervention groupparaquat group,alcohol group,control group,and long-term alcohol intervention group.The early alcohol intervention group was given alcohol first,followed by paraquat;the alcohol intervention group was given both alcohol and paraquat;the paraquat group was given paraquat;alcohol group was given alcohol;the control group was given saline;the long-term alcohol intervention group was given alcohol for 4 weeks before paraquat.After 12 h of administration,2 ml of blood was collected,and the serum was collected after centrifugation.After treatment,UPLC-MS/MS analysis was performed.Results Compared with the control group,after paraquat poisoning,lactic acid,threonine,alanine and adenine increased(P<0.05),and glutamine,pyruvate,tryptophan and phenylalanine decreased(P<0.05);in the early alcohol intervention group and the simultaneous alcohol intervention group,lactic acid became higher(P<0.01),and lactic acid decreased after long-term alcohol intervention(P<0.05);after the intervention of paraquat with alcohol,the concentration of pyruvate and tryptophan increased,and the difference was not statistically significant compared with that in the control group(P>0.05).Conclusion Compared with the early alcohol intervention group and the simultaneous alcohol intervention group,the metabolite changes caused by intervention after long-term drinking are smaller,but its mechanism needs further study.
