摘 要:目的 分析2018年和2019年上海市即食食品工厂单核细胞增生李斯特菌污染及其分子生物学特征。方法 2018年8月和2019年8月对即食食品工厂生产环节进行采样,包括食物75份和环境43份。使用脉冲场凝胶电泳(PFGE)和多位点序列分型(MLST)对分离的单核细胞增生李斯特菌进行分子分型,使用PCR对菌株进行血清群和毒力基因检测。结果 该食品厂118份样品中单核细胞增生李斯特菌检出率为16. 1%(19/118),其中原辅料检出率为33. 3%(14/42),中间产品检出率为27. 8%(5/18),香肠成品和环境样品未检出。19株分离株血清群主要为Ⅱc(1/2c,3c)和Ⅱa(1/2a,3a);分为9种PFGE带型和5种ST型。PFGE和MLST结果显示原材料和中间产品之间菌株存在克隆传播。结论 该食品厂原辅料和中间产品中存在致病性单核细胞增生李斯特菌污染,应加强原材料采购和运输过程的卫生监督,降低单核细胞增生李斯特菌对成品香肠的污染风险。Objective To analyze the contamination and molecular biological characteristics of Listeria monocytogenes in ready-to-eat food factories in Shanghai during 2018-2019.Methods Sampling was performed in the production process of ready-to-eat food factory during August 2018-August 2019,including 75 food samples and 43 environmental samples.Pulsed-field gel electrophoresis(PFGE)and multilocus sequence typing(MLST)were used to analyze the molecular typing of the strain,and PCR was used to analyze the serum group and virulence genes of the isolated strain.Results The detection rate of L.monocytogenes was 16.1%(19/118)in 118 samples,including 33.3%(14/42)in raw materials and 27.8%(5/18)in intermediate products.None of L.monocytogenes strains was isolated in sausage finished products and non-food samples.The serum groups of 19 isolates were mainly Ⅱc(1/2c,3c) and Ⅱa(1/2a,3a).There are 9 PFGE band types and 5 ST types.PFGE and MLST results showed there was clonal transmission between raw materials and intermediate products.Conclusion The contamination of pathogenic Listeria monocytogenes in raw materials and intermediate products of the food factory should be strengthened,so as to reduce the contamination risk of Listeria monocytogenes to sausage product.
