摘 要:以金银花为原材料,采用超声辅助酶法提取金银花中的多糖,在单因素试验的基础上,选用酶用量、超声温度、料液比为自变量,以多糖得率为响应值,通过响应面Box-Behnken优化金银花多糖的提取工艺。结果表明:超声辅助酶法提取金银花多糖的最佳工艺为料液比1∶48(g/mL)、超声温度50.80℃、酶用量2.56%、超声时间60 min、超声功率200 W、酶解pH值为5,在此条件下金银花多糖平均得率为54.275%。Taking Lonicera japonica as raw material,Lonicera japonica polysaccharides were extracted by ultrasonic-assisted aqueous enzymatic method.On the basis of single factor experiment,the extraction process of Lonicera japonica polysaccharides was optimized by response surface Box-Behnken design using the parameters of enzyme dosage,ultrasound temperature and solid-liquid ratio as independent variables and the yield of polysaccharides as response value.The optimal extraction process of Lonicera japonica polysaccharides by ultrasonic-assisted aqueous enzymatic method was obtained:solid-liquid ratio 1∶48(g/mL),ultrasound temperature 50.80℃,enzyme dosage2.56%,ultrasound treatment 60 min,ultrasound power 200 W,and enzymolysis pH 5.Under these conditions,the average yield of Lonicera japonica polysaccharides was 54.275%.
