摘  要:本研究以阐明阻断蚕卵滞育机制为目标,通过获得连续时间点的转录组数据,深入解析滞育过程的分子变化。采用高通量测序技术对高浓度氧、HCl处理的滞育蚕卵(以下简称"处理组")和未处理的对照组,在产后26 h、32 h和44 h取样进行转录组测序,在产后26 h到32 h阶段,处理组和对照组的共同差异基因(DEGs) 233个,占整个数量的44.5%;32 h到44 h的阶段,处理组和对照组共同DEGs仅16个,占整体数量的6.7%,但是此时处理组的共同DEGs为93个,占整体数量的38.9%。进一步对筛选的DEGs进行GO和KEGG通路分析,GO分析表明DEGs在处理组中在氧化还原功能上得到了显著富集,但是在对照组中没有得到显著富集;KEGG分析发现在产后26 h到32 h,处理组和对照组的DEGs都在溶酶体等代谢通路上调富集,但在产后32 h到44 h,处理组的蚕卵代谢通路变化相似,都在甘氨酸、丝氨酸和苏氨酸等代谢通路富集,而对照组在精氨酸和脯氨酸等代谢通路富集。综上,在产后初期,发育时间对蚕卵发育的影响大于处理的影响,然而到了后期处理的影响则逐渐突显。With the goal of elucidating the mechanism of blocking the diapause of silkworm eggs, the study was performed by obtaining transcriptomic data at consecutive time points, and in-depth analysis of molecular changes in the diapause process. Using high-throughput sequencing technology, diapause silkworm eggs treated with high concentration of oxygen and HCl(referred to as the "treatment group") and untreated controls were sampled at 26,32, and 44 h postpartum for transcriptome sequencing. The DEGs among in hyperoxia treatment, HCl treatment and control were 233 at 26 to 32 h after oviposition, accounting for 44.5% of the total number. The DEGs among in hyperoxia treatment, HCl treatment and control were 16 at 32 to 44 h after oviposition, accounting for 6.7% of the total number, whereas the DEGs in both hyperoxia and HCl treatments were 93, accounting for 38.9% of the total number. The GO and KEGG pathways of the selected DEGs were further analyzed. GO analyses showed that DEGs were significantly enriched in the redox function in the hyperoxia and HCl treatments, but not in the control.KEGG analysis showed that DEGs were significantly upregulated in the lysosomal metabolic pathway in treatments and control at 26 to 32 h after oviposition;DEGs were similar in both treatments at 32 to 44 h after oviposition, the metabolic pathways of silkworm eggs in the treatment group changed similarly. Glycine, serine and threonine were enriched in metabolic pathways, while arginine and proline were enriched in metabolic pathways.Taken together, the development time had a greater impact on the development of silkworm eggs than the treatment in the early period after oviposition, but the impact of the treatment gradually became more prominent in later period after oviposition.
