摘 要:为进一步发掘锦鲤(Cyprinus carpio)皮肤组织功能基因,以三色锦鲤墨色、红色、白色皮肤组织为研究对象,采用Illumina Hiseq 2500技术进行转录组测序和数据分析,共获得425 813 488条原始读序(reads),总碱基数为63.88 Gb。对测序所获得的reads进行组装和拼接,获得56 018条Unigenes,总长度为1 357100 421 bp,平均长度为2 380 bp。Unigenes与Nr、GO、KEGG、KOG、Pfam、Swiss-Prot和eggNO共七个数据库进行BLAST相似性比对(e-value≤10^(-5)),分别对应有19 443 (34.71%)条、7 960 (14.21%)条、10 302 (18.39%)条、12 559(22.42%)条、12 145 (21.68%)条、9 596 (17.13%)条和17 528 (31.29%)条Unigenes成功注释,20 323条Unigene至少在一个数据库中得到注释。经KEGG代谢通路分析,该转录组Unigenes参与六大类共50小类,共158份代谢通路。转录组分析还鉴定出墨色、白色、红色皮肤组织的SNP标记分别512 635个、508 711个、517 222个,转换是颠换的1.63倍。随机选取9个差异基因进行RT-qPCR验证发现,试验结果与转录谱结果一致,表明转录组测序结果是可靠的。通过研究获得了大量的三色锦鲤转录组信息资源,为研究观赏鱼皮肤功能基因、体色基因定位、遗传多样性和分子标记开发等提供了重要的基础数据。To further explore the functional genes of Koi(Cyprinus carpio) skin tissues, we used Illumina Hiseq 2500 technology to sequence and analyze the transcriptomes of the ink, red, and white Koi skin tissues. A total of 425 813 488 raw reads amounting to 63.88 Gb of sequence data were generated. The reads obtained by sequencing were assembled and spliced to obtain 56 018 Unigenes, with a total length of 1 357 100 421 bp and an average length of 2 380 bp. BLAST similarity comparison(e-value≤10^(-5)) with seven databases including Nr, GO, KEGG,KOG, Pfam, Swiss-Prot and egg NOG was conducted, corresponding to 19 443(34.71%) and 7 960(14.21%), 10 302(18.39%), 12 559(22.42%), 12 145(21.68%), 9 596(17.13%) and 17 528(31.29%) unigene successfully annotated.A total of 20 323 Unigenes were annotated in at least one database. According to the analysis of KEGG metabolic pathways, the transcriptome unigenes participates in six categories and 50 subcategories, with a total of 158 metabolic pathways. Transcriptome analysis also identified 512 635, 508 711, and 517 222 SNP markers for ink, white,and red of Koi skin tissues, respectively, and transitions were 1.63 times as large as transversions. Random selection of 9 differential genes for RT-qPCR verification found that the test results and transcript spectrum results indicated that the transcriptome sequencing results are reliable. A large number of the koi transcriptome information resources have been obtained in our research, which provided important basic data for the study of ornamental fish skin function genes, body color gene positioning, genetic diversity and molecular marker development.
