摘  要:为了研究线粒体COI基因作为DNA条形码对少鳞鳜属(Coreoperca)鱼类进行物种鉴定的可行性。采用PCR扩增与测序技术与GenBank已有序列联合分析的方法,对少鳞鳜属3种84个个体的条形码区段进行比较分析。结果表明,碱基T(U)的平均含量为30.3%,C的平均含量为27.8%,A的含量为23.1%,G的含量为18.8%。在4种碱基中,G的含量最低,其在第三密码子的使用率上也低至9.3%。少鳞鳜属3种鱼类种内平均遗传距离值为0.003 8,种间为0.146 3,种间遗传距离是种内的38倍;通过构建的系统发育树,少鳞鳜属中的分支明确,种间无交叉情况,说明种间具有明显差异,种内没有亚种的分化。研究证明,线粒体COI基因可作为条形码序列对少鳞鳜属鱼类物种鉴别的有效片段,并且还为其物种的鉴定与分类提供辅助方法。In order to study the feasibility of mitochondrial COI gene as DNA barcode for species identification of Coreoperca genus. Using the methods of PCR amplification and sequencing technology combined with the sequences of GenBank, the barcode segments of 84 individuals from three species of Coreoperca were compared and analyzed. The results showed that the average content of base T(U) was 30.3%, the average content of C was 27.8%,the content of A was 23.1%, and the content of G was 18.8%. Among the four bases, G was the lowest, and its use rate on the third codon was below 9.3%. The average genetic distance between the three species of the Coreoperca was 0.003 8, and the genetic distance between species was 0.146 3, which was 38 times more than the former;Through the construction of the phylogenetic tree, the branches in the Coreoperca genus were clear, and there was no cross between the species, indicating that there was significant difference between the species, also no subspecies differentiation within the species. Studies have shown that the mitochondrial COI gene barcode sequence can be used as an effective fragment for the species identification of the Coreoperca. It also provides an auxiliary method for the identification and classification of its species.
