摘  要:目的探讨隔山消提取物对功能性消化不良(FD)模型大鼠脑肠肽及胃肠道功能的作用机制。方法将48只雄性SD大鼠随机分为正常组(A组,生理盐水),模型组(B组,0.5 mol/L冰盐酸),多潘立酮组(C组,31.5 mg/kg多潘立酮),以及隔山消提取物低、中、高剂量组(D1组、D2组、D3组,0.5,1.0,2.0 g/kg隔山消提取物),各8只。除A组外,所有大鼠按8.0 m L/kg容量灌胃0.5 mol/L冰盐酸,同时采用不规则喂养,以构建FD大鼠模型,A组大鼠灌胃等量生理盐水。所有大鼠造模第11天后分别给予相应药物;造模第21天后,取各组大鼠小肠、胃和血液,测定小肠推进率,采用酶联免疫吸附法测定血浆中胃动素(MTL)、胃泌素(GAS)、血管活性肠肽(VIP)、肿瘤坏死因子-α(TNF-α)的水平,采用苏木精-伊红染色法观察大鼠胃窦组织的病理变化。结果与B组相比,C组、D1组、D2组、D3组大鼠的小肠推进率和血浆GAS水平均显著升高,TNF-α水平均显著降低,C组、D3组大鼠的MTL水平均显著升高,C组、D2组、D3组大鼠的VIP水平均显著降低(P<0.05或P<0.001);B组大鼠部分黏膜层细胞空泡变性,未见明显脱落、炎性细胞浸润及坏死,其余各组大鼠胃组织整体结构正常。结论隔山消提取物可通过降低FD模型大鼠血浆VIP和TNF-α水平,升高MTL和GAS水平,从而促进胃肠道运动,改善胃肠道功能。Objective To investigate the effect mechanism of the extract of Cynanchum auriculatum Radix on brain intestinal peptide and gastrointestinal function in model rats with functional dyspepsia(FD).Methods A total of 48 male SD rats were randomly divided into the normal group(group A,normal saline),the model group(group B,0.5 mol/L of glacial hydrochloric acid),the domperidone group(group C,31.5 mg/kg of domperidone),and low-,medium-and high-dose of the extract of Cynanchum auriculatum Radix groups(groups D1,D2,D3,0.5,1.0,2.0 g/kg of the extract of Cynanchum auriculatum Radix),8 rats in each group.Except for the rats in group A,other rats were given 0.5 mol/L glacial hydrochloric acid by gavage at a dose of 8.0 m L/kg,and given irregular diet to establish FD rat model.Rats in group A were given the same volume of normal saline by gavage.On the 11 th day after modeling,all rats were given corresponding drugs.On the 21 st day after modeling,the small intestine,stomach and blood of rats were taken from each group,the small intestine propulsion rate was measured,the levels of motilin(MTL),gastrin(GAS),vasoactive intestinal peptide(VIP)and tumor necrosis factor-α(TNF-α)in plasma were determined by enzyme-linked immunosorbent assay,and the pathological changes of gastric antrum tissue of rats were observed by hematoxylin-eosin(HE)staining.Results Compared with those in group B,the small intestinal propulsion rate and the levels of plasma GAS in groups C,D1,D2 and D3 were significantly increased,while the level of TNF-α in groups C,D1,D2 and D3 were significantly decreased,the levels of MTL in groups C and D3 were significantly increased,and the levels of VIP in groups C,D2 and D3 were significantly decreased(P<0.05 or P<0.001).In group B,some of the mucosa cells of rats were vacuolar degeneration,and no obvious shedding,inflammatory cell infiltration and necrosis were found.However,the overall structure of stomach tissue of rats in other groups was normal.Conclusion The extract of Cynanchum auriculatum Radix can reduce
