摘 要:目的建立测定甘草中甘草苷和甘草酸含量的替代对照品法。方法以橙皮苷为替代对照品,通过测定甘草苷和甘草酸铵相对橙皮苷的校正因子,利用相对校正因子计算甘草中甘草苷和甘草酸的含量。色谱柱为Welch Ultimate C18柱(250 mm×4.6 mm,5μm),流动相为0.1%三乙胺溶液(用磷酸调节pH至2.5)-乙腈(梯度洗脱),流速为1.0 m L/min,检测波长为276 nm(甘草苷)、284 nm(橙皮苷)、252 nm(甘草酸铵),进样量为10μL,柱温为30℃。结果甘草苷、橙皮苷、甘草酸铵的进样量分别在0.0830~1.0381μg、0.0987~1.2337μg、0.2165~2.7063μg范围内与其峰面积(r=0.9995,0.9994,0.9997)线性关系良好;甘草苷和甘草酸铵与橙皮苷的相对校正因子的平均值分别为1.0813和0.4495;甘草苷和甘草酸的平均加样回收率分别为99.62%和98.44%,RSD分别为0.87%和1.19%(n=6)。结论该方法经济快速,可行实用,准确度高,重复性好,可用于甘草中甘草苷和甘草酸的含量测定。Objective To establish a replacement method of chemical reference substance for content determination of liquiritin and glycyrrhizic acid in Glycyrrhiza uralensis.Methods Hesperidin was used as a substitute for reference substance,and the relative correction factors of liquiritin and ammonium glycyrrhizinate with hesperidin were determined to calculate the contents of liquiritin and glycyrrhizic acid in Glycyrrhiza uralensis.The chromatographic column was Welch Ultimate C18 column(250 mm×4.6 mm,5μm),the mobile phase was 0.1% triethylamine solution(p H was adjusted to 2.5 with phosphoric acid)-acetonitrile with gradient elution,the flow rate was1.0 m L/min.the detection wavelength was 276 nm for liquiritin,284 nm for hesperidin and 252 nm for ammonium glycyrrhizinate,the injection volume was 10μL,and the column temperature 30℃.Results The linear ranges of liquirtin,hesperidin and ammonium glycyrrhizinate were 0.0830-1.0381μg(r=0.9995),0.0987-1.2337μg(r=0.9994),0.2165-2.7063μg(r=0.9997),respectively.The relative correction factors of liquiritin,ammonium glycyrrhizinate with hesperidin were 1.0813 and 0.4495,respectively.The average recoveries of liquiritin and glycyrrhizic acid were 99.62%(RSD=0.87%,n=6)and 98.44%(RSD=1.19%,n=6),respectively.Conclusion The method is economical,rapid,practical,accurate and reproducible,which can be used for the content determination of liquiritin and glycyrrhizic acid in Glycyrrhiza uralensis.
