摘  要:本试验以嗜水气单胞菌CVCC-4002为宿主从鲤鱼内脏中分离得到一株噬菌体,命名为BLCC16-001,用PEG8000浓缩、超速离心获得高纯度的噬菌体进行电镜观察。测定噬菌体宿主谱、最佳感染复数(MOI)、一步生长曲线、热稳定性、pH稳定性及噬菌体体外杀菌效果。结果表明:电镜下,噬菌体BLCC16-001头部直径约60 nm,尾部长约140 nm,还能够裂解4株嗜水气单胞菌,其最佳感染复数为0.0001。噬菌体BLCC16-001潜伏期为20 min,随后进入平台期,爆发量约为1000 PFU/cell;能够耐受60℃以下的温度,超过70℃会急剧失活;对酸碱均具有一定的抵抗力,在pH为4.0~10.0时稳定;在感染复数为0.1时,3 h对宿主菌杀菌率超过90%,感染复数为1时,3 h对宿主菌杀菌率超99%。噬菌体BLCC16-001是一株宽宿主谱的能够耐受一定温度和酸碱环境的肌尾科新型烈性噬菌体,对其研究可为嗜水气单胞菌噬菌体的研究提供理论基础。Aeromonas hydrophila is a kind of conditional pathogenic bacteria,which is the most popular typical zoonotic pathogen in China.The biological characteristics of Aeromonas hydrophila isolated were studied in order to provide theoretical reference for the prevention and treatment of Aeromonas hydrophila.Using Aeromonas hydrophila CVCC-4002 as the host,a phage was isolated from carp viscera,named BLCC16-001,concentrated with PEG8000 and ultracentrifuged to obtain high-purity phage for electron microscope observation.The phage host spectrum,multiplicity of infection(MOI),one-step growth curve,thermal stability,pH stability and phage in vitro bactericidal effect were determined.The results showed that the head diameter of BLCC16-001 phage was about 60 nm,and the tail length was about 140 nm under electron microscope.It could lyse 4 strains of Aeromonas hydrophila with an optimal MOI of 0.0001.The latent period of BLCC16-001 phage was 20 min,and entered the plateau phase beyond 120 min,the burst size was about 1000 PFU/cell.It could survive below 60℃,and rapidly inactivate above 70℃;It had certain resistance to acid and alkali,and keep stable when the pH was 4.0~10.0.The bactericidal rate for the host bacteria exceeds 90%in 3 h when the MOI was 0.1,and exceeds 99%when the MOI was 1.BLCC16-001 was a new type of virulent phage of Myoviridae which could withstand certain temperature and acid-base environment with wide host spectrum.The study could provide theoretical materials for the study of phage of Aeromonas hydrophila.
