摘  要:目的:建立附子汤高效液相色谱(HPLC)指纹图谱,并结合化学模式识别比较其汤剂与配方颗粒的差异。方法:采用thermo C18色谱柱(250 mm×4.6 mm,5μm),以乙腈-0.1%磷酸水溶液为流动相,流速0.8mL·min^(-1),检测波长230 nm,柱温30℃,进样量30μL。采用"中药色谱指纹图谱相似度评价系统"考察色谱图相似度,结合系统聚类分析、主成分分析及正交偏最小二乘判别分析,综合评价汤剂(TD)、配方颗粒混冲(FGm)及配方颗粒混煎(FGd)附子汤的差异;并以变量重要性投影值(VIP)大于1.5为标准,筛选关键差异性组分。结果:建立了附子汤HPLC指纹图谱,3组样品色谱图相似度波动于0.969~0.998之间。共标定21个特征峰,均归属到各单味药,并确认了6个峰成分。与配方颗粒相比,传统汤剂缺少12号峰,二者的9、10(芍药苷)、11号峰差异较大(VIP>1.5),化学模式识别能明显将二者区分开。结论:附子汤配方颗粒和汤剂HPLC图谱有较高相似度,但仍有一定的差异,配方颗粒共煎5 min也未能提高与汤剂组的相似度。指纹图谱与化学模式识别结合可全面评价配方颗粒和汤剂的差异,并筛选出质量差异关键组分,为生产工艺优化及质量监督工作提供参考。OBJECTIVE To establish the high performance liquid chromatograph(HPLC)fingerprint of Fuzi Decoction and combine with chemical pattern recognition for comparing the differences between decoction and formula granules(FG).METHODS HPLC was performed on a Thermo C18column(250 mm×4.6 mm,5μm)with acetonitrile-0.1%phosphoric acid solution for gradient elution at aflow rate of 0.8 mL·min^(-1).The detection wavelength was 230 nm,column temperature 0℃and injection volume 30μL.The Chinese Medicine Chromatographic Fingerprint Similarity Evaluation System was employed for examining the similarity of chromatograms,along with hierarchical cluster analysis,principal component analysis and minimal orthogonality deviation Orthogonal partial least squares discriminant analysis for comprehensively evaluating the differences among traditional decoction(TD),formula granule mixed(FGm)and formula granule decocted(FGd)Fuzi decoction.And with the standard of variable importance projection value(VIP)>1.5,different key components were screenedout.RESULTS AHPLC fingerprint of Fuzi decoction was established.The similarity of three groups of sample chromatogram fluctuated between 0.969 and 0.998.Twenty-one characteristic peaks were calibrated,all of them belonged to these single medicine and 6 of them were confirmed.As compared with formula granules,TD set lacked peak 12 and obvious differences existed in the 9 th,10 th(paeoniflorin)and 11 th peaks(VIP>1.5)between them.Chemical pattern recognition could distinctly distinguish them.CONCLUSION Fuzi decoction and formula granules have different chemical compositions and 5-min co-decoction fails to improve the similarity.Combining chemical pattern recognition and fingerprint can comprehensively evaluate the differences between formula granule and decoction.And key characteristic components are screened out to guide the optimization of production process and quality supervision.
