异甘草酸镁对四氯化碳诱导肝损伤大鼠肝组织Bax、Bcl-2蛋白表达及肝纤维化指标影响

摘　　要：目的:观察异甘草酸镁注射液对四氯化碳诱导肝损伤大鼠肝组织Bax、Bcl-2蛋白表达及纤维化指标的影响。方法:50只大鼠随机分成正常对照组(NS组)、肝损伤模型组(MO组)、异甘草酸镁注射液高剂量组(H组)、异甘草酸镁注射液中剂量组(M组)、异甘草酸镁注射液低剂量组(L组),每组10只。MO组和各给药组大鼠将50%CCl4花生油溶液按照1 mL·kg^(-1),每周灌胃2次。同时,H组、M组和L组每日一次,分别腹腔注射异甘草酸镁注射液100 mg·kg^(-1)、50 mg·kg^(-1)和25 mg·kg^(-1)。6周后处死,取大鼠肝脏,观察大体形态,称重留样;取大鼠血清,测总胆红素(TBIL)、谷草转氨酶(AST)及谷丙转氨酶(ALT)水平,测透明质酸(HA)、层粘连蛋白(LN)、Ⅳ型胶原(CⅣ)和Ⅲ型前胶原肽(PC-Ⅲ)。Western blot测肝脏组织Bcl-2和Bax蛋白表达。结果:与MO组相比,异甘草酸镁注射液能够升高H组、M组大鼠体质量(P<0.01、P<0.05);降低H组肝脏指数(P<0.05);降低H组谷丙转氨酶、谷草转氨酶(P<0.05、P<0.01);降低H组LN、CIV和PC-Ⅲ(P<0.01、P<0.01、P<0.01),降低M组LN和PC-Ⅲ(P<0.01、P<0.05);下调H组Bax表达(P<0.01);上调H组Bcl-2的表达(P<0.01)。结论:对四氯化碳诱导肝损伤大鼠,异甘草酸镁注射液可下调其Bax蛋白表达,上调Bcl-2蛋白表达,改善肝纤维化指标。OBJECTIVE To observe the effects of magnesium isoglycyrrhizinate injection(MgIG)on protein expressions of Bax and Bcl-2 and fibrotic parameters in liver tissue of rats with carbon tetrachloride-induced liver injury.METHODS Fifty rats were randomly divided into five groups of NS,MO,H,M and L(n=10 each).MO and administration groups received 50%CCl4 peanut oil solution at 1 mL·kg^(-1)twice a week by gavage.At the same time,H group received MgIG 100 mg·kg^(-1)once a day and M group MgIG 50 mg·kg^(-1)once a day.L group was intraperitoneally injected with 25 mg·kg^(-1)MgIG once a day.At Week 6,the rats were sacrificed and liver samples harvested for observing general morphology and weighing and retaining the samples.The serum levels of total bilirubin(TBIL),aspertate aminotransferase(AST),alanine aminotransferase(ALT),hyaluronic acid(HA),laminin(LN),collagen typeⅣ(CⅣ)and typeⅢpre-collagen peptide(PC-Ⅲ)were recorded.Western blot was employed for measuring the protein expressions of Bcl-2 and Bax in liver tissue.RESULTS As compared with MO group,MgIG could boost body weight of groups H and M(P<0.01,P<0.05).Liver parameter decreased in group H(P<0.05).In group H,ALT and AST declined(P<0.05,P<0.01).In group H,LN,CIV and PC-Ⅲdecreased(P<0.01,P<0.01,P<0.01).In group M,LN and PC-Ⅲdiminished(P<0.01,P<0.05).Bax was down-regulated(P<0.01)while Bcl-2 up-regulated in group H(P<0.01).CONCLUSION MgIG can down-regulate the expression of Bax protein,up-regulate the expression of Bcl-2 protein and improve the hepatic fibrotic parameters in rats with liver injury induced by carbon tetrachloride.
