摘  要:目的探讨北京地区老年人KCNJ11(rs5219)和CYP2C8(rs10509681)多态性与2型糖尿病(type 2 diabetesmellitus,T2DM)的关联。方法选取2019年1—12月北京市新居里社区、北下关社区、牡丹园东里社区、大柳树社区等多个社区65岁以上老年人321例,分组为T2DM组(161例)和健康组(160例)。采用Mass ARRAY核酸质谱方法,检测KC-NJ11(rs5219)和CYP2C8(rs10509681)基因多态性,并分析其基因型频率和等位基因频率,进行比较。探讨隐性、显性或加性基因型模式与T2DM的关联。结果KCNJ11 rs5219的次要等位基因频率(minor allele frequency,MAF)在T2DM组为49.38%、健康组为39.06%,差异有统计学意义(P<0.05);KCNJ11 rs5219的隐性(CC比TC+TT,P=0.037)、显性(TT比TC+CC,P=0.027)和加性(TC比TT+CC,P=0.008)模型与T2DM有关。CYP2C8(rs10509681)的MAF在T2DM组为4.66%、健康组为1.25%,差异有统计学意义(P<0.05);CYP2C8(rs10509681)隐性(TT比CT+CC,P=0.018)模型与T2DM有关,而显性模型(CC比CT+TT,P=0.115)和加性(CT比TT+CC,P=0.058)模型与T2DM无关。结论KCNJ11(rs5219)和CYP2C8(rs10509681)基因多态性均与T2DM显著相关。Objective To explore the correlation between KCNJ11(rs5219)and CYP2C8(rs10509681)polymorphisms and type 2 diabetes mellitus(T2DM)of the elderly in Beijing.Methods A case-control study was conducted to evaluate 321 elderly people over 65 years old in Beijing.They were divided into the diabetes group(n=161)and the control group(n=160).The polymorphisms of KCNJ11(rs5219)and CYP2C8(rs10509681)genes were detected by Mass ARRAY,and their allele and genotype frequencies were analyzed and compared.The correlation between recessive,dominant or additive genotypic patterns with T2DM were studied.Results The minimum allele frequency(MAF)of KCNJ11(rs5219)was 49.38%in the diabetes group and 39.06%in the control group,and there was a significant difference between the two groups(P<0.05).The recessive(CC vs.TC+TT,P=0.037),dominant(TT vs.TC+CC,P=0.027)and additive(TC vs.TT+CC,P=0.008)models of KCNJ11(rs5219)were associated with diabetes.Similarly,MAF of CYP2C8(rs10509681)was 4.66%in the diabetes group and 1.25%in the control group,and there was significant difference between the two groups(P<0.05).The recessive(TT vs.CT+CC,P=0.018)model was associated with diabetes,while the dominant model(CC vs.CT+TT,P=0.115)and the additive model(CT vs.TT+CC,P=0.058)were not associated with diabetes.Conclusions KCNJ11(rs5219)and CYP2C8(rs10509681)gene polymorphisms are significantly associated with T2DM.
