摘  要:目的:观察南蛇藤提取物(celastrus orbiculatus ethylacetate extract,COE)对MGC803/miR-144^(+)细胞增殖和迁移能力的影响。方法:将不同浓度的COE(0 mg·L^(-1)、20 mg·L^(-1)、40 mg·L^(-1)、80 mg·L^(-1)、160 mg·L^(-1)、320 mg·L^(-1))加入对数生长期的人胃癌MGC803/miR-144^(+)细胞中,作用24 h后,MTT法检测MGC803/miR-144^(+)细胞的增殖率,根据半数抑制浓度,确定后续实验所用的药物浓度。同时设立空白质粒对照组、不加药阴性组、COE组和5-氟尿嘧啶注射液(50 mg·L^(-1))组,作用24 h后,细胞划痕实验分析迁移能力,Western Blot法检测上皮间质转化相关蛋白(N-cadherin、Vimentin及E-cadherin)的表达。结果:COE作用24 h后,MGC803/miR-144^(+)细胞的增殖能力受抑制,并呈一定的浓度依赖性(P<0.05),半数抑制浓度约为126.86 mg·L^(-1)。与对照组相比,COE可使MGC803/miR-144^(+)细胞的迁移能力受到抑制(P<0.05);可显著增加E-cadherin蛋白表达,降低N-cadherin和Vimentin蛋白表达(P<0.05)。结论:COE可抑制MGC803/miR-144^(+)细胞的增殖和迁移能力,其机制可能与上皮间质转化相关。Objective:To observe the effect of celastrus Orbiculatus ethylacetate extract(COE)on the proliferation and migration of MGC803/miR-144^(+)cells.Methods:Different concentrations of COE(0 mg·L^(-1),20 mg·L^(-1),40 mg·L^(-1),80 mg·L^(-1),160 mg·L^(-1),320 mg·L^(-1))were added to growth phase human gastric cancer MGC803/miR-144^(+)cells.After 24 hours of treatment,the MTT method was used to detect the proliferation rate of MGC803/miR-144^(+)cells,and the concentration of the drug used in subsequent experiments was determined based on the half inhibitory concentration.At the same time,control group,negative group,COE group and 5-fluorouracil injection group(50 mg·L^(-1))were set up.After 24 hours of treatment,the migration ability was analyzed by scratch test,and the expression of epithelial mesenchymal transition related proteins(N-cadherin,Vimentin and E-cadherin)was detected by Western blot.Results:After 24 hours of drug action,the proliferation of MGC803/miR-144^(+)cells was inhibited and showed a certain concentration dependence(P<0.05).The inhibitory concentration of half of the samples was 126.86 mg·L^(-1).Compared with the control group,COE inhibited the migration of MGC803/miR-144^(+)cells(P<0.05);it can significantly increase the expression of E-cadherin protein,and decrease the expression of N-cadherin and Vimentin protein(P<0.05).Conclusion:COE can inhibit the proliferation and migration of MGC803/miR-144^(+)cells,and its mechanism may be related to epithelial-mesenchymal transition.
