摘 要:目的比较尿促皮素(urocortion,UCN)同源肽与异丙肾上腺素(isoproterenol,ISO)诱导乳大鼠心肌细胞肥大的作用,并探讨与[Ca^(2+)]i瞬间变化的关系。方法采用体外培养的乳大鼠心肌细胞,分别加入不同药物处理48 h后进行指标测定。指标测定使用计算机图像分析系统检测心肌细胞体积;[3 H]亮氨酸掺入法对心肌细胞蛋白的合成进行测定;使用Lowry等法对心肌细胞蛋白含量进行检测;使用Western blot对心钠素(atrialnatriureticpeptide,ANP)表达进行测定;采用Till阳离子测定系统,以Fura-2/AM为荧光探针,观察心肌细胞[Ca^(2+)]i瞬间变化。结果UCN同源肽及ISO均能使心肌细胞体积、蛋白合成、蛋白含量和ANP表达增加(P<0.01);与其他组相比较UCNⅡ组作用最强;UCN同源肽能够使心肌细胞内[Ca^(2+)]i瞬间变化幅度及频率增高,但对静息钙无影响。结论UCN同源肽能够诱导心肌细胞肥大,其中UCNⅡ致心肌细胞肥大作用最强,其心肌细胞肥大作用可能通过改变心肌细胞内钙离子稳态来诱导。Objective To compare the effects of UCN(UCN)homologous peptide and isoproterenol(ISO)on myocardial hypertrophy in neonatal rats,and to explore their relationship with[Ca^(2+)]i transient changes.Methods Cardiomyocytes of suckling rats were cultured in vitro and treated with different drugs for 48 h.The myocardial cell volume was measured by computer image analysis system.[3H]Leucine incorporation was used to determine the synthesis of cardiomyocyte protein.Lowry and other methods were used to detect the protein content of cardiomyocytes.Western blot was used to determine the expression of ANP.The transient changes of[Ca^(2+)]i in myocardial cells were observed by the Till cation measuring system with Fura-2/AM as a fluorescent probe.Results Both UCN homologous peptide and ISO increased myocardial cell volume,protein synthesis,protein content and ANP expression(P<0.01).Compared with other groups,UCNⅡgroup had the strongest effect.UCN homologous peptides increased the amplitude and frequency of[Ca^(2+)]i transient changes in cardiomyocytes,but had no effect on resting calcium.Conclusion UCN homologous peptide can induce myocardial hypertrophy.UCNⅡhas the strongest effect on myocardial hypertrophy,which may be induced by changing the homeostasis of calcium ions in cardiomyocytes.
