摘 要:对圆齿野鸦椿枝条提取物的抑菌活性成分进行分离、纯化,结合现代波谱学技术对其结构进行鉴定,分别鉴定为(+)-(8R,7′R,8′R)-isolariciresinol-9-(6-tris-p-coumaroyl)-O-β-D-glucopyranoside(1)、(7R,8R)-threo-7,9,9′-三羟基-3,3′-二甲氧基-8-O-4′-新木脂素7-O-α-L-鼠李糖苷(2)、4-氧代芝麻素(3)、tetraketide(4)、19α-羟基熊果酸(5)、紫檀芪(6)、evofolin B(7)、豆甾醇(8).其中,除化合物4、5、7外,其他成分均为本课题组首次从野鸦椿属分离所得.抑菌活性测定结果显示,化合物4、5、7对辣椒疫霉病菌均具有中等抑制活性,其有效中浓度(EC50)值分别为1.285、0.300、0.906μmol·L^(-1).辣椒疫霉病菌经19α-羟基熊果酸处理后,菌丝干重减少,细胞膜的通透性增加,菌体内蛋白质含量增多,其保护酶(过氧化氢酶、过氧化物酶、超氧化物歧化酶)活性随着处理时间的延长呈倒"V"型变化趋势.这说明19α-羟基熊果酸对辣椒疫霉病菌具有有效抑制作用.In order to investigate the active constituents of Euscaphis konishii Hayata against Phytophthora capsici,chemical constituents extracted from the anti-fungal active part of E.konishii twig were isolated,purified and further identified by modern spectral analysis techniques.The chemicals were identified as(+)-(8R,7′R,8′R)-isolariciresinol-9-(6-tris-p-coumaroyl)-O-β-D-glucopyranoside,(7R,8R)-threo-7,9,9′-trihydroxy-3,3′-dimethoxy-8-O-4′-lignans7-O-α-L-rhamnoside,4-oxysesamin,tetraketide,pomolic acid,pterostilbene,evofolin B and stigmasterol.Except for tetraketide,pomolic acid and evofolin B,all other compounds were isolated from this genus for the first time by our research group.Anti-fungal activity testing showed that compounds tetraketide,pomolic acid and evofolin B exerted moderate inhibitory activities against P.capsici,with the EC50 value being 1.285,0.300 and 0.906μmol·L^(-1),respectively.Furthermore,hyphae dry weight,relative electrical conductivity,contents of malondialdehyde and protein,and activities of protective enzymes(CAT,POD,SOD)of the mycelia treated with pomolic acid were determined to elucidate the anti-fungal mechanism.The results showed that hyphae dry weight was decreased,permeability of cell membrane was enhanced and protein content of the mycelia increased when treated with pomolic acid.The contents of protective enzymes showed inverted V-shaped variation trends with the extension of treatment time.Therefore,pomolic acid had an effective inhibition on P.capsici,which provided a theoretical basis for the development of plant-derived fungicides.
