摘 要:目的:初步探讨柚皮素对慢性阻塞性肺疾病(COPD)大鼠模型肺功能的保护作用。方法:将40只SD大鼠分为空白组、模型组、柚皮素低剂量组和柚皮素高剂量组,每组10只大鼠。除空白组大鼠外,其余各组大鼠均联合使用脂多糖与香烟进行COPD造模。从首次使用脂多糖后第1天开始,为柚皮素低剂量组大鼠使用低剂量的柚皮素进行灌胃,为柚皮素高剂量组大鼠使用高剂量的柚皮素进行灌胃,为空白组大鼠与模型组大鼠使用等量的生理盐水进行灌胃。在末次给药24 h后,使用小动物呼吸功能测定仪检测大鼠的肺功能,收集其肺泡灌洗液进行细胞分类计数,采集其肺脏组织进行病理学检查。然后比较各组大鼠的肺功能指标、肺泡灌洗液细胞分类计数的结果和病理学检查结果。结果:1)与空白组大鼠相比,模型组大鼠的FVC、FEV1、PEF均较低,其肺泡灌洗液中性粒细胞百分比、嗜酸性粒细胞百分比、淋巴细胞百分比均较高,P<0.05;与模型组大鼠相比,柚皮素高剂量组大鼠的FVC、FEV1、PEF均较高,其肺泡灌洗液中性粒细胞百分比、嗜酸性粒细胞百分比、淋巴细胞百分比均较低,P<0.05;柚皮素低剂量组大鼠与模型组大鼠的FVC、FEV1、PEF、肺泡灌洗液中性粒细胞百分比、嗜酸性粒细胞百分比、淋巴细胞百分比相比,P>0.05。2)模型组大鼠的肺脏与支气管结构损伤严重,使用高剂量的柚皮素可减轻大鼠肺脏与支气管的损伤程度。结论:高剂量的柚皮素能减少COPD大鼠模型肺脏中中性粒细胞、嗜酸性粒细胞和淋巴细胞的数量,修复其被破坏的肺脏结构,改善其肺功能。Objective:To investigate the protective effect of naringin on lung function in rats with chronic obstructive pulmonary disease(COPD).METHODS:Forty SD rats were divided into blank group,model group,naringin low-dose group and naringin high-dose group,with 10 rats in each group.Except for the blank group,the other groups were combined with LPS and cigarettes for COPD modeling.From day 1 after the first application of LPS,rats in the naringin low-dose group were given low dose naringin by gavage,rats in the naringin high-dose group were given high dose naringin by gavage,and rats in the blank group and model group were given the same amount of normal saline by gavage.24 h after the last administration,the lung function of the rats was detected by small animal respiratory function analyzer,the alveolar lavage fluid was collected for cell classification count,and the lung tissues were collected for pathological examination.Then the lung function indexes,the results of the alloveolar lavage fluid cell classification count and the pathological examination results were compared.RESULTS:1)Compared with blank group,FVC,FEV1 and PEF in model group were lower,and the percentage of neutrophils,eosinophils and lymphocytes in alveolar lavage fluid in model group were higher,P<0.05;Compared with model group,FVC,FEV1 and PEF of naringin high-dose group were higher,and the percentage of neutrophils,eosinophils and lymphocytes in alveolar lavage fluid were lower(P<0.05).Compared with model group,FVC,FEV1,PEF,percentage of neutrophils,eosinophils and lymphocytes in naringin low-dose group(P>0.05).2)The lung and bronchial structures of rats in model group were seriously damaged,and the use of high dose naringin could reduce the degree of lung and bronchial injuries.CONCLUSIONS:High dose naringin can reduce the number of neutrophils,eosinophils and lymphocytes in the lung of COPD rats,repair the damaged lung structure and improve the lung function.
